Structural and functional dissection of Pacinian corpuscle reveals a multicellular mechanism of touch detection independent of the outer core

(Submitter supplied) Lamellated corpuscles are rapidly adapting mechanoreceptors that detect transient touch and high-frequency vibration in many vertebrates. It the prevailing model, these properties are determined by outer core, which acts as a mechanical filter that limits static and low-frequency stimuli from reaching afferent terminal—the sole site of touch detection in corpuscles. Using focused ion beam scanning electron microscopy and single corpuscle electrophysiology, we determine the detailed architecture of corpuscular components and reveal their contribution to touch detection. more...

Effects of wheat germ diet on intestinal antioxidant capacity, immunological function and gut microbiota of Sichuan white geese

(Submitter supplied) Wheat germ is known for its antioxidant, anti-inflammatory, and disease resistance properties in animals. However, its effect on the gut of Sichuan white geese remains unclear. In this study, thirty 250-day-old geese were divided into three equal groups, the control group, LWG group (21.8% wheat germ) and HWG group (43.6% wheat germ), the experiment lasted 12 weeks. We assessed various aspects of geese intestinal health, including barrier function, digestibility, antioxidant capacity, immunity, microbiota, and metabolism. more...

Whole transcriptomic analysis of liver tissue from high-intake fed compared to normally-fed lion-head geese

(Submitter supplied) Lion-head goose is the only large goose species in China, and it was one of the largest goose species in the world. Our previous study firstly reported a chromosome-level genome assembly of Lion-head goose (Anser cygnoides), a native breed in South China, through the combination of PacBio, Bionano, and Hi-C technologies. The fat content of foie gras is augmented during its preparation due to the special feeding regimen. more...

Architecture and function of lamellar cells in Meissner corpuscle

(Submitter supplied) Meissner corpuscles detect transient touch and vibration in glabrous skin, but their ultrastructure and mechanism of function are poorly understood. The Meissner corpuscle sensory core contains the terminal neurite(s) of the mechanoreceptor afferent surrounded by Schwann cell-derived lamellar cells. The afferent is thought to be the sole touch-sensing element within the corpuscle, whereas the role of lamellar cells is unclear. more...

Dosage compensation of Z sex chromosome genes in avian fibroblast cells

(Submitter supplied) Sex in birds is genetically determined, molecular mechanism of which is not well-understood. Their Z sex chromosome (chrZ) lacks whole chromosome inactivation as known for mammalian chrX. To investigate the extent of chrZ dosage compensation and its role in somatic cell’s sex specification, we used a highly-quantitative method and analyzed transcriptional activities of male and female fibroblasts from seven birds. more...

miRNA profile of duck duodenum mucosa damage caused by oxidative stress

(Submitter supplied) We deep sequenced and analyzed miRNAs using deep RNA sequencing (RNA-seq) in cage rearing and traditional breeding duck's duodenum sample of Nonghu NO.2 duck. 21 differentially expressed miRNA were identified in the duodenum. 6 miRNAs were upregulated and 15 were downregulated in the cage rearing duck's duodenum of the Nonghu NO.2 duck compared to their expression in the control group. These findings provided insights into the expression profiles of miRNAs in duck duodenum, and deepened our understanding of miRNAs in oxidative injury of duck.

Genome-wide circRNA profiling reveals that circGAS2-2 are abundantly expressed and regulates myoblast proliferation in indigenous Chinese duck breed

(Submitter supplied) The objective of this study was to identify candidate circular RNAs associated with duck muscle development. circRNA sequencing analysis was employed using female breast muscle samples embryo stage 13th day (E13) and embryo stage 19th day (E19). RNA-seq data and validation experiment in duck myoblast cells showed that circGAS2-2 significantly differential expressed between E13 and E19 group.

(Submitter supplied) The transcription expressions of gout goslings in renal tissues was significantly different from that of healthy goslings. With P <0.05 and |log2(fold change)| >1 as the selection standards, 1749 differential expression genes were found. Among these, 501 genes were up-regulated and 1248 genes were down-regulated in the gout group. The gene ontology (GO) search revealed many significant terms, including chemokine activity, T cell receptor binding, antigen binding, immune response, G-protein coupled receptor signaling pathway, chemokine-mediated signaling pathway, inflammatory response and so on. more...

Transcriptome Sequencing to Identify Important Genes and lncRNAs Regulating Abdominal Fat Deposition in Ducks

(Submitter supplied) Abdominal fat deposition is an important trait in meat-producing ducks. F2 generations of 304 Cherry Valley and Runzhou Crested White ducks were studied to identify genes and lncRNAs affecting abdominal fat deposition. RNA sequencing was used to study abdominal fat tissue of four ducks each with high or low abdominal fat rates. In all, 336 upregulated and 297 downregu-lated mRNAs, and 95 upregulated and 119 downregulated lncRNAs were identified. more...

Transcriptomic analysis of the duckling liver mRNA methylome following infection by the virulent and attenuated Hepatitis A virus type 1 strains

(Submitter supplied) Duck hepatitis A virus (DHAV) modulates host physiology and gene expression. The RNA modification N6-methyladenosine (m6A) modulates mRNA fate and thus affects many biological processes in mammals. Neither modifications on mRNAs have been reported in avian species. Here, we used immunoblot analysis and LC-MS/MS to show the presence of m6A modification in ducking liver, and a decreased m6A modification by virulent DHAV-1 (CH) strain when compared to the attenuated vaccine DHAV-1 (CK). more...

RNAseq data of duck liver after feeding choline chloride

(Submitter supplied) We aim to study the unusual TMA metabolism mechanism of ducks, and further explore the hidden reasons that led to the weakening TMA metabolism ability. To achieve this, transcriptome, proteome, and metagenome analyses were integrated based on the constructed duck populations with high TMA metabolism ability and low TMA metabolism ability. In addition, further experiments were followed to validate the hypothesis on the limited flavin-containing monooxygenase 3 (FMO3) metabolism ability of ducks. more...

Single cell atlas for mammals, reptiles and birds

(Submitter supplied) The availability of viral entry factors is a prerequisite for the cross-species transmission of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Large-scale single-cell screening on animal cells is a powerful tool to reveal the expression patterns of viral entry genes for different hosts. But such exploration for SARS-CoV-2 remained limited. Here, we presented the broadest pan-species single-nucleus RNA sequencing study to date, covering 11 representative species in pets (cat, dog, hamster, lizard), livestock (goat, rabbit), poultry (duck, pigeon) and wildlife (pangolin, tiger, deer), from which we investigated the co-expression of ACE2 and TMPRSS2. more...
