Increased viral tolerance mediates by antiviral RNA interference in bat cells

(Submitter supplied) Bats harbor highly virulent viruses that can infect other mammals, including humans, posing questions about their immune tolerance mechanisms. Bat cells employ multiple strategies to limit virus replication and virus-induced immunopathology, but the coexistence of bats and fatal viruses remains poorly understood. Here, we investigated the antiviral RNA interference (RNAi) pathway in bat cells and discovered that they have an enhanced antiviral RNAi response, producing canonical viral small interfering RNAs (vsiRNAs) upon Sindbis virus (SINV) infection that were missing in human cells. more...

Transcriptomic profiling of bat PakiT03 cells with immune stimulation

(Submitter supplied) To examine innate immune signalling, IFN baseline and IRF-regulation, we profile Pteropus alecto bat kidney epithelial cells (PakiT03) with Deep NGS coverage. Clonal cell lines were also isolated with CrispR KO for IRF1, IRF3, IRF7, IFNAR2 or IFNAR2/IRF7 double KO.

Transcriptomic profiling of bat immune responses (P. alecto)

(Submitter supplied) To examine the fundamental immunity in bats, particularly the status of their innate immune system in the basal healthy state, we profile Pteropus alecto bat tissue with Deep NGS coverage. This is coupled to a paired experiment where bats were stimulated in vivo with various PRR ligands to activate immune pathways.

Transcriptional response to type I IFN in bat P. alecto cells

(Submitter supplied) Bats are reservoirs of a number of highly pathogenic human viruses, yet they remain relatively asymptomatic during infection. Whether this viral resistance is due to a unique immune system is unknown. An evolutionarily conserved feature of vertebrate antiviral immunity is the interferon (IFN) response, which triggers cellular defenses through interferon-stimulated gene (ISG) expression. While bats encode an intact IFN system, global ISG expression patterns in bat cells are not well characterized. more...
