Mussel Immunochip 1.0

(Submitter supplied) Immunochip 1.0 includes 1820 spotted oligonucleotides representing putative immune-related transcripts of M. galloprovincialis. The probe ID also indicates the related transcript sequence available at http://mussel.cribi.unipd.it. For instance, MGO_00175 relates to MGC00175 in Mytibase, a dynamic knowledgebase of expressed mussel sequences described in BMC Genomics 2009 Feb 9;10:72. Probes were designed with OligoArray 2.1. more...

Performance of Immunochip 1.0 with hemolymph samples collected at 3 and 48 hours from Vibrio-challenged mussels

(Submitter supplied) We aimed to exemplify early and late transcriptional response of M. galloprovincialis to live Vibrio cells. Hemolymph was collected at 3 and 48 hours after the injection of 10 to 7 cells Vibrio splendidus LGP32 into the posterior adductor muscle. Hemolymph samples were similarly collected from paired control mussels injected with PBS-NaCl. The purified RNAs were successfully amplified, labelled and competitively hybridized to the new mussel oligoarray Immunochip 1.0. more...

Revealing Mytilus galloprovincialis transcriptomic profile during ontogeny

(Submitter supplied) Mediterranean mussels are a worldwide spread bivalve species with extraordinary biological success. One of the reasons of this success could be the reproduction strategy of bivalves, characterized by the presence of trochophore larvae. Larval development in bivalves has been a topic of raising interest in the scientific community but it deserves much more attention. The principal objective of this work was to study the transcriptomic profile of the ontogeny of M. more...

Gene expression profiling of native mussels from Venice lagoon area

(Submitter supplied) We examined gene expression profiling of native mussels that were sampled in early summer 2003 from sites of the Venice lagoon area known to be differently affected by chemical pollution: Sites 1 and 2 close to the industrial district of Marghera and Site 3 close to the Lido lagoon outlet. Site 4, a current mussel farm located offshore, has been chosen as source of reference targets for microarray hybridizations. more...

Gene expression profiling of mussels treated with two chemical mixtures

(Submitter supplied) We examined gene expression profiling of mussels treated with two chemical mixtures composed by heavy metals or organic compounds, both including genotoxic agents (cadmium, benzo[a]pyrene, 1-nitro/amino-pyrene) and other contaminants (cupper, mercury, fluoranthene, TCDD). For digestive gland one RNA sample from control mussels and one from each treatment group (7 mussels) were competitively hybridized on the MytArray 1.0. more...

Tissue specificity of gene expression profiling from mussels in normal conditions.

(Submitter supplied) We examined tissue specificity of gene expression in control mussels. Total RNA was isolated from gills (G), digestive gland (DG), tissues involved in contraction/motility (FML: foot/adductor muscles/ligaments) and reproduction (GM: gonads and mantle) producing four tissue-specific samples. Total RNA was reverse-transcribed and labeled using a Cy3-dCTP and Cy5-dCTP direct incorporation. Each sample was hybridized in competition with the total pool (equal RNA amounts from G, DG, GM and FML) and the relative abundance of each gene was measured by directly comparing fluorescent signals for each probe. more...

Mussel MytArray 1.0

(Submitter supplied) MytArray 1.0 is a cDNA platform, that consists of 1,712 different clones obtained from systematic sequencing of multiple Mytilus galloprovincialis tissues cDNA library and 46 unrelated controls (ESTs from Coffea and bacterial species) printed in duplicate and twice per slide. This library was produced using a strategy that allows the selection of a short 3’-end region of mRNAs and the tagging of each transcript with an unique probe. more...

Heat and copper induced mRNA changes in mussels

(Submitter supplied) Transcriptional profiling of the digestive gland tissue of female mussel Mytilus galloprovincialis exposed to copper along with a temperature gradient Background: Global warming is a major factor that may affect biological organization, especially in marine ecosystems and in coastal areas that are particularly subject to anthropogenic pollution. We evaluated the effects of simultaneous changes in temperature and copper concentrations on lysosomal membrane stability of the blue mussel Mytilus galloprovincialis (Lam.). more...
