Glioblastoma pseudopalisading cells

Analysis of pseudopalisading cells obtained by laser capture microdissection from glioblastoma tumors. The presence of pseudopalisading cells is associated with aggressive malignant gliomas. Results identify putative prognostic markers for glioblastoma.

Gene expression profiling in glioblastoma and immunohistochemical evaluation of IGFBP-2 and CDC20

(Submitter supplied) The different clinical behavior of low and high grade gliomas and the chance to develop novel selective agents that specifically target tumor-associated proteins in gliomas stimulate the research of molecules playing a role in glioma progression. Gene expression profiling using microarrays allows the study at the same time of the expression patterns of thousands of genes in tumor cells. In the present study microarrays with about 20,000 genes have been employed to discover the gene expression profile in 39 glial neoplasias (28 glioblastomas (GBM) and 11 low grade gliomas, namely 4 oligodendrogliomas, 5 pilocytic astrocytomas (PA), 2 fibrillary astrocytomas (FA)). more...

5`aza-dC demethylation of three short term cultured glioblastomas

(Submitter supplied) Glioblastoma, the most aggressive and least treatable form of malignant glioma, is the most common human brain tumor. Although many regions of allelic loss occur in glioblastomas, relatively few tumor suppressor genes have been found mutated at such loci. To address the possibility that epigenetic alterations are an alternative means of glioblastoma gene inactivation, we coupled pharmacological manipulation of methylation with gene profiling to identify potential methylation-regulated, tumor-related genes. more...

DNA demethylation effect on glioblastoma cultures

Analysis of short-term cultured glioblastoma cells treated with 5-aza-dC to induce DNA demethylation. Glioblastoma cells derived from 3 independent primary glioblastomas. Results provide insight into the role of epigenetic alterations in glioblastoma gene inactivation.

Clinical and Molecular Characteristics of Congenital Glioblastoma Multiforme

(Submitter supplied) Congenital glioblastoma multiforme (cGBM) historically has been considered an aggressive tumor of infancy requiring extensive chemotherapy to achieve cure. We report on 4 patients at our institution with cGBMs who were treated with surgery and chemotherapy (carboplatin and etoposide every 21 days for 2-6 cycles). Four of four patients are progression free at a median time of 27.5 months (22-103 months). more...

Differential gene expression in astrocytic tumours

(Submitter supplied) This study aims at investigating differential gene expression in human astrocytic tumours of grades I, II, III and IV. A total of 65 tumours were assessed using the Affymetrix U133A GeneChip. The study aims not only at discovering the main expression differences between astrocytic tumours of distinct histological grades but also wishes to correlate these findings with the known histopathological hallmarks of astrocytoma progression. more...

Isolation and characterization of cancer-associated fibroblast-like cells in the glioblastoma microenvironment

(Submitter supplied) Glioblastoma (GB) is a highly infiltrative tumor recurring in 90% of cases within a few centimeters of the resection cavity, even in cases of complete tumor resection and adjuvant chemo/radiotherapy. This observation highlights the importance of understanding this special zone of brain tissue surrounding the tumor. It is becoming clear that the non neoplastic stromal compartment of most solid cancers plays an active role in tumor proliferation, invasion and metastasis. more...
