Regulation of signaling genes by TGFbeta during entry into dauer diapause in C. elegans

(Submitter supplied) BACKGROUND: When resources are scant, C. elegans larvae arrest as long-lived dauers under the control of insulin/IGF- and TGFbeta-related signaling pathways. : We have identified genes that show different levels of expression in a comparison of wild-type L2 or L3 larvae (non-dauer) to TGFbeta mutants at similar developmental stages undergoing dauer formation. Many insulin/IGF pathway and other known dauer regulatory genes have changes in expression that suggest strong positive feedback by the TGFbeta pathway. more...

Gene expression C. elegans age-1 mutants with neuron- or intestine-restricted age-1 activity

(Submitter supplied) The goal of this experiment was to identify transcripts regulated non-autonomously by age-1 activity in neurons or intestine. Comparisons of gene expression were made between age-1(mg44) null adults and CY251 (age-1(mg44); bvIs2) and CY262 (age-1(mg44); bvIs1); all were relative to wildtype (N2). The integrated transgenes, bvIs2 and bvIs1, carry the age-1 cDNA expressed from the neuronal ric-19 promoter (bvIs2) or intestinal ges-1 promoter (bvIs1). more...

daf-16/FoxO isoform-specific deletion mutants

(Submitter supplied) FoxO transcription factors promote longevity across taxa. How they do so is poorly understood. In the nematode Caenorhabditis elegans, the A- and F-isoforms of the FoxO transcription factor DAF-16 extend life span in the context of reduced DAF-2 insulin-like growth factor receptor (IGFR) signaling. To elucidate the mechanistic basis for DAF-16/FoxO-dependent life span extension, we performed an integrative analysis of isoform-specific daf-16/FoxO mutants. more...

Transcriptome analysis of hpl-2(tm1489) vs wild-type worms at mixed embryonic and third larval stages

(Submitter supplied) Epigenetic factors guide cell fate decisions and can stabilize development by buffering environmental variation. HP1 proteins have a well-characterized role in heterochromatin packaging and gene regulation, while their function in organismal development is less well understood. Here we used genome-wide expression profiling to assess novel functions of the C. elegans HP1 homologue HPL-2 at specific developmental stages

daf-16/FOXO blocks adult cell fate in Caenorhabditis elegans dauer larvae via lin-41/TRIM71

(Submitter supplied) Many tissue-specific stem cells maintain the ability to produce multiple cell types during long periods of non-division, or quiescence. FOXO transcription factors promote quiescence and stem cell maintenance, but the mechanisms by which FOXO proteins promote multipotency during quiescence are still emerging. The single FOXO ortholog in C. elegans, daf-16, promotes entry into a quiescent and stress-resistant larval stage called dauer in response to adverse environmental cues. more...

Life span-extending effects of royal jelly and its related substances on the nematode Caenorhabditis elegans

(Submitter supplied) One of the most important issues in the study of aging is to discover compounds with longevity-promoting activity and to unravel their underlying mechanisms. Queen honey bees are continuously fed royal jelly (RJ), and they live more than 10 times longer than hive workers, derived from the same diploid genome, which are fed it only for a short period of time during their larval stages. Therefore, RJ is likely to contain longevity-promoting agents for queens. more...

Activation of DAF-16/FOXO by reactive oxygen species promotes longevity in long- lived mitochondrial mutants in C. elegans

(Submitter supplied) Mild deficits in mitochondrial function have been shown to increase lifespan in multiple species including worms, flies and mice. Here, we study three C. elegans mitochondrial mutants (clk-1, isp-1 and nuo-6) to identify overlapping genetic pathways that contribute to their longevity. We find that genes regulated by the FOXO transcription factor DAF-16 are upregulated in all three strains, and that the transcriptional changes present in these worms overlap significantly with the long-lived insulin-IGF1 signaling pathway mutant daf-2. more...

RNAseq of dauer-forming C. elegans L2d in daf-2 and daf-2;daf-42

(Submitter supplied) Under adverse environmental conditions, nematodes arrest into dauer, an alternative developmental stage for diapause. Dauers endure unfavorable environment and interact with host animals to access favorable environments, thus playing a critical role in the survival of both free-living and parasitic nematodes. Here, we discovered that in Caenorhabditis elegans, daf-42 is essential for development into dauer stage, as the null mutant shows lethal phenotype during dauer entry. more...

Insulin-like Signaling Determines Survival During Stress via Post Transcriptional Mechanisms in C. elegans.

(Submitter supplied) The insulin-like signaling (ILS) pathway regulates metabolism and is known to modulate adult lifespan in C. elegans. Altered stress responses and resistance to a wide range of stressors are also associated with changes in ILS and contribute to enhanced longevity. The transcription factors DAF-16 and HSF-1 are key effectors of the longevity phenotype. We demonstrate that increased intrinsic thermotolerance, due to lower ILS, is not dependent on stress induced HSF-1 transcriptional responses but instead requires active protein translation. more...
