Retinal gene expression in chicks during imposed myopic defocus

(Submitter supplied) The retina plays an important regulatory role in ocular growth. To screen for new retinal candidate genes that could be involved in the inhibition of ocular growth, we used chick microarrays to analyze the changes in retinal mRNA expression after myopic defocus was imposed by positive lens-wear. Chicks were raised under a 12h-light/12h-dark cycle (light-onset: 8:00 am and light-offset: 8:00 pm) with unrestricted access to water and food. more...

Microarray analysis of changes in gene expression within the amacrine cell layer of chicks following optical defocus

(Submitter supplied) Ocular growth is regulated locally by signals produced in the retina that ultimately act on the growth of the scleral tissue. Consequently, a number of studies have investigated changes in retinal gene expression during manipulation of ocular growth in an attempt to elucidate the biochemical pathways underlying eye growth. However, due to the highly heterogenous nature of the retina, important changes in gene expression can be masked. more...

Retinal gene expression in Egr-1 knock-out mice during development (p30 and p42)

(Submitter supplied) In chicks, the avian homologue of the early growth response protein-1 (ZENK) has been shown to be increased in a special cell type of the retina, the glucagonergic amacrine cells, under conditions that lead to a reduction in eye growth (myopic defocus, recovery of myopia) and decreased under conditions that enhance ocular growth (hyperopic defocus, form-deprivation). The investigation of Egr-1 knock-out mice showed that homozygous knock-out mice with no functional Egr-1 protein developed relative axial myopia at the age of 42 and 56 days, compared to heterozygous- and wildtype Egr-1 knock-out mice. more...

Egr-1 deficiency effect on retina during eye development

Analysis of retinas of Egr-1 deficient animals at post-natal days 30 and 42. 42 day-old Egr-1 knockout animals have longer eyes and a more myopic refractive error compared to wild-types. Results provide insight into the molecular mechanisms regulating axial eye growth.

Image defocus and altered retinal gene expression in chicks

(Submitter supplied) Because refractive development is governed largely by the retina, we analyzed the retinal transcriptome in chicks wearing a spectacle lens, a well-established means to induce refractive errors, to identify gene expression alterations and to develop novel mechanistic hypotheses about refractive development.

Image defocus effect on retina/retinal pigment epithelium: time course

Analysis of retina and retinal pigment epithelium (RPE) from 1 week-old chicks wearing a unilateral spectacle lens of + or 15 diopters for 6 hours or 3 days. The spectacle lens induces refractive errors. Results provide insight into molecular mechanisms underlying refractive development.

Chick myopia

(Submitter supplied) One-day old white Leghorn chicks were housed in brooders with a 12 hr light:dark cycle, using General Electric chroma 50 fluorescent lighting with irradiance of approximately 50μW/cm2 at chick eye level. They received Purina Chick Chow food and water ad libitum. At one week of age and at the onset of the light phase, the chicks were anesthetized with inhalation ether, and a unilateral translucent white plastic goggle was glued to the periorbital feathers to induce ipsilateral form-deprivation myopia, alternating between the left or right eye. more...

Comparison of Saffron and Photobiomodulation on the light damaged rat retina.

(Submitter supplied) Aim: To identify the genes and non-coding RNAs (ncRNAs) involved in the neuroprotective actions of a dietary anti-oxidant (saffron) and of photobiomodulation. Methods: We used a previously published assay of photoreceptor damage, in which albino Sprague Dawley rats raised in dim cyclic illumination (12h 5 lux, 12h darkness) are challenged by 24h exposure to bright (1,000 lux) light. Experimental groups were protected against light damage by pretreatment with dietary saffron (1mg/kg/day for 21d) or photobiomodulation (10 J/cm2 at the eye, daily for 5d). more...

Daily Patterns of Retinal and Choroidal Gene Expression and Circadian Biology in Myopia Pathogenesis

(Submitter supplied) We investigated diurnal changes in gene expression in the retina and choroid at the onset of experimental myopia. We induced visual form deprivation myopia in young chicks, and isolated retinal and choroidal tissues separately in form- deprived and contralateral control eyes every 4 hours over a single 24-hour period.

Gene Expression Analysis of the Rat Retina during Maturation

(Submitter supplied) Recently we reported that the rat inner retina undergoes significant functional changes during maturation. Aiming to gain knowledge on additional aspects of retinal development and maturation, we used the microarray system to monitor gene expression patterns in the rat retina at ages 5, 11, and 20 weeks. The analysis revealed the expression of many well-documented retinal genes as well as a high number of non–annotated genes. more...

Effects of Aging and Anatomic Location on Gene Expression in Human Retina

(Submitter supplied) Objective: To determine the effects of age and topographic location on gene expression in human neural retina. Methods: Macular and peripheral neural retina RNA were isolated from human donor eyes for DNA microarray and quantitative RTPCR analyses. Results: Total RNA from human donor retina preserved integrity. Hierarchic clustering analysis demonstrates the gene expression profiles of young, old, macula and peripheral retina cluster into four distinct groups. more...
