Differential gene expression in Danio rerio during optic nerve regeneration

(Submitter supplied) To gain a better understanding of the factors necessary for successful CNS regeneration, a temporal analysis of the changes in gene expression in the eye caused by optic nerve injury was conducted. Dual color oligonucleotide microarrays were used to compare total RNA harvested from the eyes of sham-operated and optic nerve-injured fish at 3, 24, and 168 hours following surgery. Optic nerve injured fish are compared to sham-operated fish in order to eliminate gene expression due to non-neuronal damage and inflammatory response. more...

Gene Expression Profiles of Intact and Regenerating Zebrafish Retina

(Submitter supplied) Purpose: Investigate the molecular determinants of retinal regeneration in adult vertebrates by analyzing the gene expression profiles of control and post-lesion retina of adult zebrafish, a system that regenerates following injury. Methods: Gene expression profiles of zebrafish retina and brain were determined with DNA microarray, RT-PCR, and real-time quantitative PCR analyses. Damaged retinas and their corresponding controls were analyzed 2-5 days post-lesion (acute injury condition) or 14 d post-lesion (cell regeneration condition). more...

temporal profiling of retinal transcriptome regulation after IONT and IONC

(Submitter supplied) retinal ganglion cells die after optic nerve injury, either crush or transection. The molecular causesunderlying this degeneration are largely unkwon the purpose of this job is to find which (if any) gene regulation triggers RGC death with the final goal of design neuroprotective protocols Keywords: time course

Retina response to optic nerve injury: time course

Temporal analysis of retinas of animals following intraorbital optic nerve transection (IONT) or intraorbital nerve crush (IONC). Optic nerve injury, which is more severe with IONT than IONC, triggers retinal ganglion cell death. Results provide insight into the molecular basis of this degeneration.

Optic nerve crush induces spatial and temporal gene expression patterns in retina of BALB/cJ mice

(Submitter supplied) Central nervous system (CNS) trauma and neurodegenerative disorders trigger a cascade of cellular and molecular events resulting in neuronal apoptosis and regenerative failure. The pathogenic mechanisms and gene expression changes associated with these detrimental events can be effectively studied using a rodent optic nerve crush (ONC) model. The purpose of this study was to use a mouse ONC model to: (a) evaluate changes in retina gene expression, (b) identify neurodegenerative pathogenic pathways and (c) discover potential new therapeutic targets. more...

Zebrafish heart regeneration

(Submitter supplied) Ischemic cardiopathy is the leading cause of death in the world, for which efficient regenerative therapy is not currently available. In mammals, after a myocardial infarction episode, the damaged myocardium is replaced by scar tissue featuring collagen deposition and tissue remodelling with negligible cardiomyocyte proliferation. Zebrafish, in contrast, display an extensive regenerative capacity as they are able to restore completely lost cardiac tissue after partial ventricular amputation. more...

Gene expression timecourse in zebrafish whole eye in response to optic nerve crush

(Submitter supplied) It is well-established that neurons in the adult mammalian central nervous system are terminally differentiated and, if injured, will be unable to regenerate their connections. In contrast to mammals, zebrafish and other teleosts display a robust neuroregenerative response. Following optic nerve crush (ONX), retinal ganglion cells (RGC) regrow their axons to synapse with topographically correct targets in the optic tectum, such that vision is restored in ~21 days. more...
