Human esophageal adenocarcinomas

(Submitter supplied) The incidence of esophageal and junctional adenocarcinoma has increased 6 fold in the last 30 years and 5 year survival remains less than 14%. Most patients present with advanced disease and current staging is limited in its ability to predict survival. We aimed to generate and validate a molecular prognostic signature for esophageal adenocarcinoma. Gene expression profiling was performed and the resulting signatures correlated with clinical features for 91 snap frozen esophageal and junctional resection specimens. more...

Integrated analysis of genome-wide copy number and matched expression data reveals novel genes in esophageal adenocarcinoma

(Submitter supplied) The incidence of esophageal and junctional adenocarcinoma has increased 6-fold in the west in the last 30 years and 5 year survival remains <14%. We aimed to characterize genome-wide aberrations in esophageal adenocarcinoma to further understand disease pathogenesis and ultimately identify groups with differential survivals with implications for clinical management. Oligo-array-based high-resolution analysis of copy number changes in 89 fresh frozen esophageal adenocarcinoma resection sections with long-term clinical follow-up data was performed. more...

The discovery and validation of ten-gene prognostic classifier in gastric cancer by whole genome expression profiling

(Submitter supplied) The aim of this study is to identify and verify a prognosis set that is able to distinguish patient overall survival status based on whole genome expression profiling. Using Illumina HumanWG-6 beadarray, we analyzed the expression of 47,296 transcripts in two groups of gastric cancer patients who all underwent surgical resection. Thirty-nine of 46 samples in group one were used as a training set to discover candidates for overall survival prediction by using semi-supervised method, resulting in a panel of 10 genes as prognosis markers. more...

Genomic and transcriptomic alterations associated with drug vulnerabilities and prognosis in adenocarcinoma at the gastroesophageal junction

(Submitter supplied) Adenocarcinoma at the gastroesophageal junction (ACGEJ) has dismal clinical outcomes and there are currently few specific effective therapies because of limited knowledge on its genomic and transcriptomic alterations. The present study investigates genomic and transcriptomic changes in ACGEJ from Chinese patients and analyzes their drug vulnerabilities and associations with the survival time. Here we show that the major genomic changes of Chinese ACGEJ patients are chromosome instability promoted tumorigenic focal copy-number variations and COSMIC Signature 17-featured single nucleotide variations. more...

Correlation of Survival with Gene Expression in Oesophageal Cancer

(Submitter supplied) We evaluated and correlated gene expression profiles of pre-treatment endoscopic biopsies in a pre-defined patient population of resectable oesophageal adenocarcinoma who had received neoadjuvant platinum and fluoropyrimidine based chemotherapy with clinical outcome, the primary end-point being overall survival. At the time of pre-treatment staging EUS, tumour biopsies were collected (approximately 1.5 mm3). more...
