Immune-activation as a therapeutic direction for patients with high-risk ovarian cancer based on gene expression signature (1)

(Submitter supplied) The Japanese Serous Ovarian Cancer Study Group Advanced-stage ovarian cancer is one of the most lethal gynecologic malignancies. To improve prognosis of patients with ovarian cancers, a predictive biomarkers leading to personalized treatments are required. In this large-scale cross-platform study of six microarray datasets consisting of 1054 ovarian cancer patients, we developed a novel risk classification system based on a 126-gene expression signature for predicting overall survival by applying elastic net7 and 10-fold cross validation to a Japanese dataset A (n = 260). more...

A Gene Expression Signature to Predict Metastasis and Survival in Non-small Cell Lung Cancer

(Submitter supplied) Background: Current histopathological methods are inadequate for predicting outcome and recurrence in patients with non-small cell lung carcinoma (NSCLC) after surgery. In this study, we investigated the use of gene expression signatures to predict outcome and metastasis in lung cancer patients. Methods: Gene expression was studied by microarray and the real-time reverse transcriptase polymerase chain reaction (RT-PCR) in normal and lung tumor tissue of 188 NSCLC patients who underwent surgical resection. more...

caArray_cho-00156: Gene Expression in Ovarian Cancer Reflects Both Morphology and Biological Behavior

(Submitter supplied) Biologically and clinically meaningful tumor classification schemes have long been sought. Some malignant epithelial neoplasms, such as those in the thyroid and endometrium, exhibit more than one pattern of differentiation, each associated with distinctive clinical features and treatments. In other tissues, all carcinomas, regardless of morphological type, are treated as though they represent a single disease. more...

A signature predicting poor prognosis in gastric and ovarian cancer represents a coordinated macrophage and stromal-response.

(Submitter supplied) Genome wide mRNA expression profiling of 94 gastric tumours derived from Australian based cohort was performed. . From this data we identified a cluster of co-expressed genes termed the stromal response cluster which almost perfectly differentiates tumor from its non-malignant gastric tissue and hence can be regarded as a highly tumor-specific gene expression signature. We show that these genes are consistently co-expressed across a range of independent gastric datasets as well as other cancer types suggesting a conserved functional role in cancer.

Search for a gene-expression signature of breast cancer local recurrence in young women

(Submitter supplied) Kreike et al, have constructed a gene-expression (GE) profile predictive for local recurrence (LR) after breast-conserving treatment (BCT) from a series of 165 patients . This study aimed to test this signature both internally (cross-platform) and externally on a independent series and to further explore the search for a GE signature of breast cancer LR in young women.

MicroRNA profiling of advanced serous ovarian carcinoma

(Submitter supplied) Objectives: MicroRNAs (miRNAs) are a class of small non-coding RNAs that negatively regulate gene expression primarily through post-transcriptional modification. We tested the hypothesis that miRNA expression is associated with overall survival in advanced ovarian cancer. Methods: Cases included newly diagnosed patients with stage III or IV serous ovarian cancer. RNA from a training set of 62 cases was hybridized to an miRNA microarray containing 470 mature human transcripts. more...

A Gene Signature Predicting for Survival in Suboptimally Debulked Patients with Ovarian Cancer

(Submitter supplied) To identify a prognostic gene signature accounting for the distinct clinical outcomes in ovarian cancer patients Despite the existence of morphologically indistinguishable disease, patients with advanced ovarian tumors display a broad range of survival end points. We hypothesize that gene expression profiling can identify a prognostic signature accounting for these distinct clinical outcomes. To resolve survival-associated loci, gene expression profiling was completed for an extensive set of 185(90 optimal/95 suboptimal) primary ovarian tumors using the Affymetrix human U133A microarray. more...

Gene expression profiles associated with ITGBL1 overexpression

(Submitter supplied) In our previous microarray study we identified two subgroups of high-grade serous ovarian cancers with distinct gene expression and different survival. Among differentially expressed genes was an Integrin beta-like 1 (ITGBL1), coding for a poorly characterized protein comprised of ten EGF-like repeats. In this study we investigated ITGBL1 influence on ovarian cancer cells phenotype. Using various functional assays we found that ITGBL1 overexpression affected cellular adhesion, migration and invasiveness, while it had no effect on proliferation rate and the cell cycle. more...

Gene expression profiling in ovarian cancer

(Submitter supplied) The introduction of microarray techniques to cancer research brought great expectations for finding biomarkers that would improve patients’ treatment; however, the results of such studies are poorly reproducible and critical analyses of these methods are rare. In this study, we examined global gene expression in 97 ovarian cancer samples. Also, validation of results by quantitative RT-PCR was performed on 30 additional ovarian cancer samples. more...

Survival Related Profile, Pathways and Transcription Factors in Ovarian Cancer

(Submitter supplied) To date, a variety of studies have employed gene expression profiling to classify ovarian carcinomas in clinically relevant subtypes. These studies provided valuable first clues to molecular changes in ovarian cancer that might be exploited in new treatment strategies. However, most studies were of relatively limited size and the number of overlapping genes in the identified profiles was minimal. Although identification of gene expression profiles associated with clinically relevant subtypes in ovarian cancer is important, knowledge is now emerging rapidly on how genes interact in pathways, networks and complexes; this allows us to unravel those cellular pathways determining the biological behavior of ovarian cancer, that might be successfully targeted with drugs. more...

OCaMIR a non-invasive early diagnostic test for ovarian cancer: A multi-cohort retrospective and prospective analysis

(Submitter supplied) Owing to the lack of effective screening tools and early detection biomarkers, ovarian cancer (OC) still remains as a deadly disease with highest mortality among other gynecological cancers. So far there have been no attempts to discover biomarkers using early stage OC patients. MicroRNAs (miRNAs) have been recognized as great tool to develop non-invasive biomarkers in various cancers including ovarian cancer.

Gene expression patterns of chemoresistant and chemosensitive serous epithelial ovarian tumors with possible predictive value in response to initial chemotherapy

(Submitter supplied) Chemotherapy (CT) resistance in ovarian cancer is broad and encompasses diverse, unrelated drugs, suggesting more than one mechanism of resistance. We aimed to analyze the gene expression patterns in primary serous epithelial ovarian cancer (EOC) samples displaying different responses to first-line CT in an attempt to identify specific molecular signatures associated with response to CT. Initially, the expression profiles of 15 chemoresistant serous EOC tumors [time to recurrence (TTR) ≤6 months] and 10 chemosensitive serous EOC tumors (TTR ≥30 months) were independently analyzed which allowed the identification of specific sets of differentially expressed genes that might be functionally implicated in the evolution of the chemoresistant or the chemosensitive phenotype. more...

A gene expression prognostic signature for overall survival in patients with high-grade serous ovarian cancer

(Submitter supplied) Expression of 276 genes was associated with OS at a false discovery rate (FDR) of < 0.05 in covariate-adjusted single gene analyses. The top five genes were TAP1, ZFHX4, CXCL9, FBN1, and PTGER3 (p ≪ 0.001). The best performing signature included 101 genes and for each SD difference in the gene expression score conferred a greater than two-fold increase in risk of death (HR = 2.35 [2.02, 2.71]; p ≪ 0.001). more...
