Alterations in bronchial airway microRNA expression for lung cancer detection

(Submitter supplied) We have previously shown that gene-expression alterations in cytologically normal appearing bronchial epithelial cells can be used as a biomarker for lung cancer detection in smokers (Whitney et al., BMC Medical Genomics 2015; Silvestri et al., NEJM 2015). In this study, we have established that there are also alterations in bronchial microRNA-expression of smokers with lung cancer. Importantly, the performance of an existing bronchial mRNA-biomarker has been improved by integrating microRNA with mRNA expression.

Characterizing the molecular spatial and temporal field of injury in early stage smoker non-small cell lung cancer patients after definitive surgery by expression profiling

(Submitter supplied) Gene expression alterations in response to cigarette smoke have been characterized in normal-appearing bronchial epithelium of healthy smokers and it has been suggested that adjacent histologically normal tissue display tumor-associated molecular abnormalities. We sought to delineate the spatial and temporal molecular lung field of injury in smoker early stage non-small cell lung cancer (NSCLC) patients (n=19) who were accrued into a surveillance clinical trial for annual follow-up and bronchoscopies within one year after definitive surgery.

Gene expression profiling of the adjacent airway field cancerization in early stage NSCLC

(Submitter supplied) Previous work has shown that lung tumors and normal-appearing adjacent lung tissues share specific abnormalities that may be highly pertinent to the pathogenesis of lung cancer. However, the global and molecular adjacent airway field cancerization in non-small cell lung cancer (NSCLC) has not been characterized before. We sought to understand the transcriptomic architecture of the adjacent airway field canerization, in conjunction with tumors, to gain additional insights into the lung cancer biology and oncogenesis.

miR-134/487b/655 Cluster regulates TGF-β-induced epithelial-mesenchymal transition and drug resistance to Gefitinib by targeting MAGI2 in lung adenocarcinoma cells.

(Submitter supplied) Epithelial-mesenchymal transition (EMT) has recently been recognized as a key element of cell invasion, migration, metastasis, and drug resistance in several types of cancer, including non-small cell lung cancer (NSCLC). Our aim was to clarify microRNA (miRNA) -related mechanisms underlying EMT followed by acquired resistance to epidermal growth factor receptor tyrosine-kinase inhibitor (EGFR-TKI) in NSCLC. more...

Temporal RNA-sequencing of cytologically-normal mouse airway epithelia during tobacco carcinogen-associated lung tumorigenesis

(Submitter supplied) Smoking perpetuates in cytologically-normal airways a molecular “field of injury” that is pertinent to lung cancer and early detection. The evolution of airway field changes prior to lung cancer onset is poorly understood largely due to the long latency of lung malignancy in smokers. Here we studied airway expression changes prior to lung cancer onset in mice with knockout of the Gprc5a gene and tobacco carcinogen (nicotine-specific nitrosamine ketone; NNK) exposure and that develop the most common type of lung cancer, lung adenocarcinoma (LUAD). more...

MicroRNA expression data from high and low invasive A549 human lung adenocarcinoma cells

(Submitter supplied) The major cause of cancer-related deaths in lung cancer patients is commonly attributed to its ability to metastasize to distant organs such as the bone, brain, liver and adrenal glands. The dysregulation in miRNA expression is believed to contribute toward the initiation and progression of metastasis through their capability to regulate target genes. In this study, two NSCLC sub-cell lines possessing opposing migration and invasion properties were established using serial transwell invasion assays, followed by miRNA microarray to obtain the overview of miRNA expression as well as identify important playes in metastasis.
