Microarray analysis of gene expression profiles in Map2k1+/-Map2k2+/- and Map2k1flox/-Map2k2+/-Tg+/Sox2Cre placentas

(Submitter supplied) The mammalian genome contains two ERK/MAP kinase kinase genes, Map2k1 and Map2k2, encoding dual-specificity kinases responsible for ERK/MAP kinase activation. Loss of Map2k1 function in mouse causes embryonic lethality due to placental defects, while Map2k2 mutant mice survive with a normal lifespan. The Map2k1 mutation interferes with the growth of the labyrinthine region of the placenta and its vascularization. more...

Transcription factor TFAP2C regulates major programs required for murine fetal germ cell maintenance and haploinsufficiency predisposes to teratomas in male mice

(Submitter supplied) Maintenance and maturation of primordial germ cells is controlled by complex genetic and epigenetic cascades, and disturbances in this network lead to either infertility or malignant aberration. Transcription factor Tcfap2c / TFAP2C has been described to be essential for primordial germ cell maintenance and to be upregulated in several human germ cell cancers. Using global gene expression profiling, we identified genes deregulated upon loss of Tcfap2c in primordial germ cell-like cells. more...

Transcription factor Tcfap2c deficiency effect on primordial germ cells

Analysis of embryonic stem cells (ESCs) depleted for transcription factor Tcfap2c and PGC-like cells (PGCLCs) derived from the Tcfap2c-deficient ESCs. TFAP2C is a member of the activator protein-2 (AP-2) family. Results provide insight into the role of TFAP2C in germ cell fate.

AKT1-FOXO4 Axis Regulates Hemochorial Placentation [Foxo4-KD-rTSC]

(Submitter supplied) AKT1 is a serine/threonine kinase implicated in fetal, placental, and postnatal growth. In this study, we investigated roles for AKT1 in placental development using a genome-edited/loss-of-function rat model. Both heterozygous and homozygous Akt1 mutant rats were viable and fertile. Disruption of AKT1 resulted in placental, fetal, and postnatal growth restriction. Akt1 null placentas showed deficits in both junctional zone and labyrinth zone size and their ability to adapt to a physiological stressor. more...

Cardiac samples from OTT1 null/null and OTT1 null/wt embryonic mice

(Submitter supplied) The infant leukemia-associated gene, Ott1(Rbm15), has broad regulatory effects within the murine hematopoiesis. However, germline Ott1 deletion results in fetal demise prior to E10.5, indicating additional developmental requirements for Ott1. The spen gene family, to which Ott1 belongs, has a transcriptional activation/repression domain and RNA recognition motifs, and in Drosophila has a significant role in the development of the head and thorax. more...

The imprinted Phlda2 gene modulates a major endocrine compartment of the placenta to regulate placental demands for maternal resources

(Submitter supplied) Recent work suggests that imprinted genes may regulate the signalling function of the placenta by modulating the size of the endocrine compartment. Our work provides in vivo evidence that this hypothesis is well founded. Affymetrix Mouse Microarray analysis of E16.5 placneta was used to demonstrate gene expression changes in the mutant placenta.

GATA2/3-TFAP2A/C transcription factor network couples human pluripotent stem cell differentiation to trophectoderm with repression of pluripotency

(Submitter supplied) To elucidate the molecular basis of BMP4-induced differentiation of human pluripotent stem cells (PSCs) toward progeny with trophectoderm characteristics, we produced transcriptome, epigenome H3K4me3, H3K27me3, and CpG methylation maps of trophoblast progenitors, purified using the surface marker APA. We combined them with the temporally resolved transcriptome of the preprogenitor phase and of single APA+ cells. more...

Unraveling the role of human TSC-specific TFs [RNA-Seq]

(Submitter supplied) Orchestrated actions of tissue-specific enhancers and transcription factors (TFs) govern the development of the placenta which is an essential organ to support the growth of the fetus during pregnancy. However, human trophoblast stem cells (TSCs)-specific enhancers, TFs, and the mechanisms by which TFs modulate placental development are poorly understood. Here we investigated enhancers, super-enhancers (SE), SE-associated genes in human TSC using RNA-seq and ChIP-seq to interrogate the roles of SE-TFs in human TSCs.

Gene expression data from AP-2γ silenced MCF-7 cells

(Submitter supplied) Overexpression of the AP-2γ transcription factor in breast tumours has been identified as an independent predictor of poor outcome and failure of hormone therapy, even in ER positive, ErbB2 negative tumours; markers of a more favourable prognosis. To understand further the role of AP-2γ in breast carcinoma, we have used an RNA interference and gene expression profiling strategy using the MCF-7 cell line as a model for ER positive, ErbB2 negative tumours with AP-2γ overexpression. more...

Altered Gene Expression in Placentae from a Mouse Model of Diabetic Pregnancy

(Submitter supplied) AIMS/HYPOTHESIS: Pregnancies complicated by diabetes have a higher risk of adverse outcomes for mothers and children, including predisposition to disease later in life, such as metabolic syndrome and hypertension. We hypothesized that adverse outcomes from diabetic pregnancies may be linked to compromised placental function. Our goal in this study was to identify cellular and molecular abnormalities in diabetic placenta. more...

Usp22 controls multiple signaling pathways that are essential for vasculature formation in the mouse placenta

(Submitter supplied) Usp22, a component of the SAGA complex, is over expressed in highly aggressive cancers, but the normal functions of this deubiquitinase are not well defined. We determined that loss of Usp22 in mice results in embryonic lethality due to defects in extra-embryonic placental tissues and failure to establish proper vascular interactions with the maternal circulatory system. These phenotypes arise from abnormal gene expression patterns that reflect defective kinase signaling, including TGFβ and several receptor tyrosine kinase (RTK) pathways. more...
