CodY of Streptococcus pneumoniae: link between nutritional gene regulation and virulence

(Submitter supplied) CodY is a nutritional regulator mainly involved in amino acid metabolism. It has been extensively studied in Bacillis subtilis and Lactococcus lactis. We investigated the role of CodY in gene regulation and virulence of the human pathogen Streptococcus pneumoniae. We constructed a codY-mutant and examined the effect on gene and protein expression by microarray and 2D DIGE analysis. The pneumococcal CodY-regulon was found to consist predominantly of genes involved in amino acid metabolism, but also several other cellular processes, such as carbon metabolism and iron uptake. more...

Global transcriptomic response of modified Bacillus anthracis over expressing protective antigen

(Submitter supplied) In this study we focused on understanding the effect of over expression of recombinant protective antigen (rPA) on the Bacillus anthracis metabolism and genes expression with the purpose of improving the production of this recombinant toxin in particular and recombinant proteins in general. To achieve this goal controlled growth of the B. anthracis ames strain transformed with pYS5 (PA producer) plasmid encoding protective antigen and the corresponding empty vector pSW4 (non-producer) were performed and protein production, bacterial growth characteristics and gene transcription pattern were analyzed. more...

Integration of metabolism and virulence by Clostridium difficile CodY

(Submitter supplied) CodY, a global regulatory protein that monitors the nutrient sufficiency of the environment by responding to the intracellular levels of GTP and the branched-chain amino acids, was previously shown to be a potent repressor of toxin gene expression in Clostridium difficile during growth in rich medium. In the intestinal tract, such derepression of toxin synthesis may lead to destruction of epithelial cells and the liberation of potential nutrients for the bacterium. more...

Strand-specific RNA-seq reveals ordered patterns of sense and antisense transcription in Bacillus anthracis

(Submitter supplied) Although genome-wide transcriptional analysis has been used for many years to study bacterial gene expression, many aspects of the bacterial transcriptome remain undefined. A prominent example is antisense transcription, which has been observed in a number of bacteria, though the function of antisense transcripts, and their distribution across the bacterial genome, is still unclear. Single-stranded RNA-seq results revealed a widespread and non-random pattern of antisense transcription covering more than two-thirds of the B. more...

B. anthracis transcriptome analysis using Illumina and SOLiD sequencing

(Submitter supplied) Although gene expression has been studied in bacteria for decades, many aspects of the bacterial transcriptome remain poorly understood. Transcript structure, operon linkages, and absolute abundance information all provide valuable insights into gene function and regulation, but none has ever been determined on a genome-wide scale for any bacterium. Here we report the first comprehensive and unbiased definition of a bacterial transcriptome under various growth conditions. more...

Transcriptome RNA Sequencing Data Sets of Bacillus anthracis Vollum DhtrA and Parental Isogenic Wild-Type Strains under Oxidative Stress Conditions

(Submitter supplied) The High Temperature Requirement (HtrA) chaperone/protease is involved in the stress-response of the anthrax-causing pathogen Bacillus anthracis. Resilience to oxidative stress is essential for manifestation of B. anthracis pathogenicity.
