Expression data from wild type mice and REDD1 null mice following acute aerobic exercise

(Submitter supplied) Regulated in DNA Damage and Development 1 (REDD1) is a stress induced protein whose expression is highly induced in skeletal muscle following a single bout of aerobic exercise. However, the role of this induction is unknown We used microarrays to detail the change in gene signature within skeletal muscle following a single bout of aerobic exercise in wild type and REDD1 null mice

Effects of testosterone on dexamethasone-induced changes in gene expression in gastrocnemius muscles from male rats

(Submitter supplied) Glucocorticoids are a well recognized and common cause of muscle atrophy. Glucocorticoid-induced atrophy can be prevented by testosterone, but the molecular mechanisms underlying such protection have not been described. Thus, the global effects of testosterone on dexamethasone-induced changes in gene expression were evaluated in rat gastrocnemius muscle using Affymetrix 230_2 DNA microarrays. Gene expression was analyzed after 7 days administration of dexamethasone, dexamethasone plus testosterone, or vehicle. more...

Transcriptomic analysis of porcine muscle Longissimus dorsi (LD) exposed to a single bout of endurance exercise

(Submitter supplied) Acute physical exercise elicits changes in gene expression in skeletal muscles to promote metabolic changes and to repair exercise-induced muscle injuries. Here, we investigated the impact of a single bout of running exercise until exhaustion on global transcriptional profiles in porcine skeletal muscles. Using a combined microarray and candidate gene approach, we identified a suite of genes that are differentially expressed in muscles during post-exercise recovery. more...

REDD1 (regulated in development and DNA damage response 1) dissociates therapeutic and adverse effects of topical steroids in skin

(Submitter supplied) Even though cutaneous atrophy is the major adverse effect of topical glucocorticoids, its molecular mechanisms are poorly understood. We found that glucocorticoids strongly increased the expression of REDD1 (regulated in development and DNA damage response 1), a stress-inducible inhibitor of mTOR, in mouse and human epidermis. We found that REDD1 knockout animals are partially resistant to glucocorticoid-induced epidermal and subcutaneous adipose atrophy which correlated with the protection of CD34+ follicular epithelial stem cells as well as p63+ keratinocyte progenitors in REDD1 knockout epidermis during chronic steroid treatment. more...

α7β1 Integrin Regulation of Gene Transcription in Skeletal Muscle Following an Acute Bout of Eccentric Exercise

(Submitter supplied) The objective of this study was to understand the mechanisms behind the contribution of the alpha7BX2 integrin to improvements and growth in skeletal muscle post exercise. We utilized global gene profiling to identify novel signaling pathways and patterns of gene expression involved in this adaptation process, and found the alpha7BX2 integrin initiates transcription of genes that allow for protection from stress and modulation of protein synthesis.

Skeletal Muscle Response

(Submitter supplied) To improve our understanding of the genetic reprogramming during the early phase of skeletal adaptation in response to endurance training, we employed cDNA microarray analysis to examine changes in mRNA expression in mouse plantaris muscle after a single bout voluntary running. Control untrained mice were compared with with R3h, R12h, P3h, P6h, P12h, P24h and R4w mice. We conclude that a single bout of voluntary running is sufficient to induce active remodeling processes, including cell proliferation, by a coordinated regulation of the genome in response to signals elicited from increased neuromuscular activity. more...

Caltech 16K cDNA Mouse Array

(Submitter supplied) The 16K mouse cDNAs probes used in this microarray were designed using sequence information gathered from RIKEN FANTOM cDNA library. Probes can be 0.5 to 3 kb in length where about 60% of the probes represent full length transcripts. The PCR products were inkjet-printed on the array by Agilent Technologies (Palo Alto, CA).
