Gene signature profiles in respiratory epithelium infected with nontuberculous mycobacteria

(Submitter supplied) The incidence of pulmonary nontuberculous mycobacterial (PNTM) disease is increasing, but host susceptibility factors are not fully understood. We infected air-liquid interface (ALI) primary respiratory epithelial cell cultures with Mycobacterium avium complex (MAC) or Mycobacterium abscessus (MAB) and performed transcriptome sequencing (RNA-Seq) to identify relevant gene expression differences. We used cells from 4 different donors in order to try to obtain generalizable data. more...

A genome-wide screen in macrophages defines host genes regulating the uptake of Mycobacterium abscessus

(Submitter supplied) The initial interactions between a host cell and a pathogen can dictate disease outcomes and are important targets for host-directed therapies. Mycobacterium abscessus (Mab) is a highly antibiotic resistant, rapidly growing non-tuberculous mycobacterium that infects patients with chronic lung diseases, like cystic fibrosis. Mab can infect host immune cells, such as macrophages, which contribute to its pathogenesis. more...

High-throughput RNA-sequencing of human macrophages infected with Mycobacterium abscessus Smooth and Rough variants

(Submitter supplied) Mycobacterium abscessus is an emerging pathogen causing pulmonary infections in those with inflammatory lung disorders, such as Cystic Fibrosis (CF), and is associated with the highest fatality rate among rapidly growing mycobacteria (RGM). Phenotypically, MAB manifests as either a Smooth (MAB-S) or a Rough (MAB-R) morphotype, which differ in their levels of cell wall glycopeptidolipids (GPLs) and in their pathogenicity in vivo. more...

Gene expression profiling of the Tlr2 mutant of zebrafish embryos at 4 days post infection of M. marinum

Human pluripotent stem cell-derived macrophages host Mycobacterium abscessus infection

(Submitter supplied) Human macrophages are a natural host of many mycobacterium species, including Mycobacterium abscessus (M. abscessus), an emerging pathogen affecting patients with lung diseases and immunocompromised individuals. There are few available treatments and the search for effective antibiotics against M. abscessus has been hindered by the lack of a tractable in vitro intracellular model of infection. Here, we established a reliable model for M. more...

Human Cytomegalovirus-Induced Interleukin-10 Production Promotes the Proliferation of Mycobacterium massiliense in Macrophages

(Submitter supplied) We profiled the transcriptome of HCMV-, HCMV/M. mass- and M. mass-infected macrophages. Comparison of hierarchical clustering with the 880 differentially expressed genes (DEGs) between the HCMV-infected and control macrophages revealed that M. mass co-infection did not globally change the transcriptome of HCMV-infected macrophages. Low immune responsiveness upon M. mass challenge was due to the existence of HCMV, which was clearly shown in the gene ontology (GO) analysis. more...

Single cell RNA sequencing of lung myeloid cells from aged (20-22 months old) mice treated with Isotype or anti-IFNAR1 antibody for 1 day or 1 week

(Submitter supplied) Monocytes and neutrophils are an important subset of myeloid cells in the lung. However, little is known about how aging affects these lung cells, and the role of Type I interferon signaling. Here, we treat aged (20-22 months old) mice with 500ug anti-IFNAR1 antibody (Clone MAR1-5A3 from BioXcell) intraperitoneally (i.p.) once or for 1 week (total 4 i.p. injections, one every other day) and sorted CD45+ CD11b+ Gr1+ cells from the lung the day after the last injection. more...
