Hemogen /BRG1 cooperativity modulates promoter and enhancer activation during erythropoiesis [RNA-seq]

(Submitter supplied) To gain insight into hemogen function during human erythropoiesis, RNA-seq was performed in different type of erythroid cells, such as WT and hemogen KD CD34+ cells, WT and hemogen KD HUDEP2 cells, WT and hemogen KO K562 cells. Notably, depletion of hemogen in these human erythroid cells significantly reduced the expression of genes associated with heme and hemoglobin synthesis, such as ALAS2, HMBS, GYPA, EPOR, and HBB, supporting a positive role of hemogen in erythroid maturation. more...

BRG1 contributes to the global chromatin accessibility revealed by its acute depletion

(Submitter supplied) DNase I hypersensitivity (DHS) of chromatin indicates the presence of important regulatory elements of transcription such as enhancers and promoters and plays critical roles for their activities. However, the mechanisms that control the global DHS have not been fully understood. In this report, we show that acute depletion of BRG1 by the auxin-dependent degron system, the essential ATPase subunit of the mammalian chromatin remodeling SWI/SNF-like BAF complexes, severely compromised genome-wide DHSs of chromatin, while the traditional conditional deletion of the Brg1 gene using Cre/Flox system led to only very modest changes of DHSs. more...

Role of SWI/SNF in acute leukemia maintenance and enhancer-mediated Myc regulation (RNA-seq)

(Submitter supplied) Cancer cells frequently depend on chromatin regulatory activities to maintain a malignant phenotype. Here, we show that leukemia cells require the mammalian SWI/SNF chromatin remodeling complex for their survival and aberrant self-renewal potential. While Brg1, an ATPase subunit of SWI/SNF, is known to suppress tumor formation in several cancer types, we found that leukemia cells instead rely on Brg1 to support their oncogenic transcriptional program, which includes Myc as one of its key targets. more...

Dynamic BRG1 Recruitment During T Helper Differentiation And Activation Reveals Distal Regulatory Elements

(Submitter supplied) We found binding of the remodeling protein BRG1 was programmed by lineage and activation signals. BRG1 binding was positively correlated with gene activity at protein-coding and miRNA genes. BRG1 binding was found at promoters and distal regions, including known and novel distal regulatory elements. Distal BRG1 binding correlated with expression, and novel distal sites possessed enhancer activity, suggesting a general role for BRG1 in long-distance gene regulation. more...
