Sex-Dependent Changes of miRNA Levels in the Hippocampus of Adrenalectomized Rats Following Acute Corticosterone Administration

(Submitter supplied) We explored sex-biased effects of the primary stress glucocorticoid hormone corticosterone on the miRNA expression profile in the rat hippocampus. Adult adrenalectomized (ADX) female and male rats received a single corticosterone (10 mg/kg) or vehicle injection and after 6 h, hippocampi were collected for miRNA, mRNA and Western blot analyses. miRNA profiling microarrays showed a basal sex-biased miRNA profile in ADX rat hippocampi. more...

Hippocampal transcriptome analysis of glucocorticoid enhanced memory formation [RNA-seq]

(Submitter supplied) Aim: We investigated whether glucocorticoid effects on memory are associated with genomic interactions between the GR and pCREB in the hippocampus. Methods: Adult male rats received 3 minutes of object exploration training or no training either with a 3.0mg.kg post-training corticosterone or vehicle injection to study the molecular effects underlying enhanced memory formation. Hippocampi were dissected 3 hours after injection and snap-frozen. more...

Glucocorticoid-Dependent Hippocampal Transcriptome in Male Rats: Pathway-Specific Alterations with Aging

(Submitter supplied) Although glucocorticoids (GCs) are known to exert numerous effects in the hippocampus, their chronic regulatory functions remain poorly understood. Moreover, evidence is inconsistent regarding the longstanding hypothesis that chronic GC exposure promotes brain aging/Alzheimer's disease. Here, we adrenalectomized male F344 rats at 15-months-of-age, maintained them for 3 months with implanted corticosterone (CORT) pellets producing low or intermediate (glucocorticoid-receptor (GR)-activating) blood levels of CORT, and performed microarray/pathway analyses in hippocampal CA1. more...

Corticosterone effect on hippocampus: dose response

Analysis of hippocampal CA1 from adrenalectomized F344 males maintained for 3 months with implanted corticosterone (CORT) pellets to produce low or intermediate (glucocorticoid receptor-activating) blood levels of CORT. Results provide insight into the effects of chronic CORT on the hippocampus.

Gene expression changes in mouse hippocampus after overnight exposure to corticosterone

(Submitter supplied) The aim of the experiment was to test the effect of elevated level of corticosterone on hippocampal transcriptome after 12 hours of treatment during active phase to mimic the effect of a day-long stress. Additionally, we also tested the decay time of transcriptomic response during resting period when the level of glucocorticoids returns to the baseline. Our study shows that transcriptomic responses to glucocorticoids are heterogenous in terms of the decay time with some genes displaying persistent changes in expression during 9 hours of rest.
