Acute rotavirus infection: peripheral blood mononuclear cells

Expression profiling of peripheral blood mononuclear cells (PBMCs) from children with acute rotavirus diarrhea. Rotavirus is the most common cause of severe diarrhea in children.

Rotavirus activates B but impairs T lymphocytes

(Submitter supplied) Rotavirus infection is the single most important cause of severe diarrhea in young children worldwide. We used Affymetrix Human U95Av2 high density oligonucleotide arrays to compare gene expression profiles in peripheral blood mononuclear cells (PBMC) of 10 children with acute rotavirus diarrhea and 8 age-matched healthy children. We also examined patterns of gene expression in 5 convalescent-phase PBMC samples from rotavirus patients. more...

Human PBMNCs: Control vs. Patients with Kawasaki disease

(Submitter supplied) Genes that were consistently up-regulated in KD patients compared with healthy controls, and that showed over 2.0 or 3.0-fold differences by the comparison between the two groups in the mean expression levels were selected.

Normal blood cell subtypes

(Submitter supplied) We used antibodies coupled to magnetic beads (MACS) to purify cell types out of whole blood from healthy donors (CD4+ T cells, CD8+ T cells, and B cells). We also profiled whole blood (collected in Pax tubes) and Peripheral Blood Mononuclear cells obtained by Ficoll-Paque based separation. RNA was extracted using Trizol, amplified linearly twice, labeled with Cy5 and co-hybridized with amplified Stratagene human reference (labeled with Cy3). more...

Immune cell subsets and their gene expression profiles from human PBMC isolated by Vacutainer Cell Preparation Tube and standard density gradient

(Submitter supplied) High quality genetic material is an essential pre-requisite when analyzing gene expression using microarray technology. Peripheral blood mononuclear cells (PBMC) are frequently used for genomic analyses, but several factors can affect the integrity of nucleic acids prior to their extraction, including the methods of PBMC collection and isolation. In this study, we compared the Ficoll-Paque density gradient centrifugation and BD Vacutainer cell preparation tube (CPT) protocols to determine if either method offered a distinct advantage in preparation of PBMC-derived immune cell subsets for their use in gene expression analysis. more...

Transcriptomes of whole blood and PBMC in chickens.

(Submitter supplied) The gene expression profiles of chicken whole blood (WB) and peripheral blood mononuclear cells (PBMC) have been characterized and compared by microarray analysis. This study highlights the respective advantages of each blood cell subset and show that whole blood is more suitable for large scale studies and allows for the exploration of biological processes which are so far poorly investigated.

Gene expression pattern in Caco-2 cells following rotavirus infection

(Submitter supplied) Rotaviruses are recognized as the leading cause of severe dehydrating diarrhea in infants and young children worldwide. Preventive and therapeutic strategies are urgently needed to fight this pathogen. In tissue culture and in vivo, rotavirus induces structural and functional alterations in the host cell. In order to better understand the molecular mechanisms involved in the events after rotavirus infection, we identified host cellular genes whose mRNA levels changed after infection. more...
