5-aza-2’-deoxycytidine effect on colorectal cancer cell lines

Analysis of colorectal cancer (CRC) cell lines treated with 5-aza-2’-deoxycytidine (5-aza-dC). 5-aza-dC, a DNA methylation inhibitor, was used to reactivate silenced genes in the CRC cell lines. Results provide insight into the molecular mechanisms underlying epigenetic alterations in CRC.

Expression profiling of HCC

(Submitter supplied) 61 human HCCs were analyzed for genome-wide gene expression. Samples were collected at two sites in Germany, Heidelberg (HD) and Hannover (N). The Heidelberg Collection include 40 independent HCC: 19 liver resections and 17 explant liver specimen (4 not determined); median age at surgery was 57 years (range, 16-78), and the male/female ratio was 3:1. Methylation-silenced genes in colorectal cancer cell lines

(Submitter supplied) Using an oligonucleotide array, we undertook a genome-wide search for genes upregulated following treatment with a demethylating agent in two CRC cell lines. Promoter methylation status was determined in 12 CRC cell lines and 11 CRC tissues. After the treatment, 350 genes were upregulated 1.5 fold or more. Six genes (PAGE-5, VCX, MAEL, GAGED2, UCHL1, and GAGE7), which contained putative 5'CpG islands in their promoter regions, were confirmed to be silenced in CRC cell lines. more...

Aberrant global methylation patterns affect the molecular pathogenesis and prognosis of human multiple myeloma
