Fra-1 deficient lung response to fibrotic agent bleomycin

Analysis of lungs from Fra-1 deficient mutants 5 days following intratracheal administration of bleomycin to induce lung fibrosis. Fra-1 is a b-ZIP transcription factor. Results provide insight into the molecular mechanisms by which Fra-1 protects against bleomycin-induced lung injury.

Comparative analysis of gene expression in Fra-1+/+ and Fra-1-/- mice lung

(Submitter supplied) We hypothesized that gene expression in lungs of Fra-1+/+ and Fra-1-/- mice are divergent thus contributing fibrosis. More specifically, Fra-1-/- mice are increased susceptible to fibrosis. In order to test these hypotheses at the gene expression level, we utilized microarray analysis to examine transcriptional differences between Fra-1+/+ and Fra-1-/- mice at early time point.

Identification of Fra-1/AP-1 binding sites in macrophages

(Submitter supplied) The polarization and activation of macrophages are controlled synergistically by transcription factors such as NF-κB and AP-1 transcription factor members. Surprisingly, little is known about the role of the AP-1 member, Fra-1, during macrophage responses. To determine the full spectrum of Fra-1 binding sites in macrophages, chromatin immunoprecipitation sequencing analysis was performed in macrophages pooling down for the transcription factor Fra-1.

RNA sequencing of lung tissue from Fra-2 transgenic mouse (Fra-2tg) and wild-type littermates

(Submitter supplied) Fra-2tg mice develop spontaneous lung fibrosis. We carried out this sequencing with tissue from young animals before they develop the tissue fibrosis, to explore the early gene expression changes that might contribute to the phenotype.

Expression data from mouse lung treated with saline or bleomycin

(Submitter supplied) Lung fibrosis represents the final common pathway of a variety of lung injuries, and it is characterized, independently of etiology, by the expansion of the fibroblast/myofibroblast population and by the abnormal accumulation of extracellular matrix (ECM) replacing normal functional parenchyma. In general, there is no effective therapy, and currently the only helpful available treatment for advanced lung fibrosis is transplantation. more...

Insights into the invasiveness of triple negative breast cancer from genome-wide profiling of transcription factor AP-1 (expression)

(Submitter supplied) Triple negative breast cancer (TNBC) is an aggressive clinical phenotype, and accounts for 15% to 20% of all breast cancers. The molecular determinants of malignant cell behaviors in TNBC remain largely unknown. We find that the AP-1 transcription factor component, Fra-1, is overexpressed in basal-like breast tumors, and its expression level has high prognostic significance. Depletion of Fra-1 or its heterodimeric partner c-Jun inhibits the proliferative and invasive phenotypes in TNBC cells. more...

Collagen Dynamics and Transcriptional Profiling in Bleomycin-induced Pulmonary Fibrosis

(Submitter supplied) Collagen deposition is a key process during idiopathic pulmonary fibrosis (IPF); however, little is known about the dynamics of collagen formation during disease development. Tissue samples of early stages of human disease are not readily available and it is difficult to identify changes in collagen content, since standard collagen analysis does not distinguish between 'old' and 'new' collagen. Therefore, the current study aimed to (i) investigate the dynamics of new collagen formation in mice using bleomycin-induced lung fibrosis in which newly synthesized collagen was labelled with deuterated water and (ii) use this information to identify genes and processes correlated to new collagen formation from gene expression analysis. more...

The AP-1 Transcription Factor c-Jun Prevents Stress-Imposed Maladaptive Remodeling of the Heart

(Submitter supplied) Systemic hypertension increases cardiac workload and subsequently induces signaling networks in heart that underlie myocyte growth (hypertrophic response) through expansion of sarcomeres with the aim to increase contractility. However, conditions of increased workload can induce both adaptive and maladaptive growth of heart muscle. Previous studies implicate two members of the AP-1 transcription factor family, junD and fra-1, in regulation of heart growth during hypertrophic response. more...

Gene expression in fra-2 transgenic mice

(Submitter supplied) Microarray analysis of Fra-2 tg female and male mice at 8 weeks of their disease development give an indication of possible differentially regulated genes. Wildtype mice serve as a healthy control. The mRNA was obtained from lung homogenate of the mice (n=4/group for all). There was no treatment applied, we investigate the baseline differences.
