Evidence of a role for CutRS and actinorhodin in the secretion stress response in Streptomyces coelicolor M145

(Submitter supplied) CutRS was the first two-component system to be identified in Streptomyces species and is highly conserved in this genus. It was reported >25 years ago that deletion of cutRS increases the production of the antibiotic actinorhodin in Streptomyces coelicolor but despite this early work the function of CutRS has remained enigmatic until now. Here we show that deletion of cutRS upregulates the production of the actinorhodin biosynthetic enzymes up to 260-fold in the cutRS mutant, explaining the increase in actinorhodin production. more...

Transcriptome profiles of Streptomyces coelicolor A3(2) wild type treated with kanamycin and iron

(Submitter supplied) We determine genes that responsible for iron induced resistance to kanamycin in Streptomyces coelicolor. Iron acts as a inducing agent for resistance to bactericidal antibiotics with concentration dependent manner. Identification of iron-dependent differentially expressed genes in wild-type by RNA-seq identified more than 100 genes. This series encompasses the RNA-seq data of our study.

(Submitter supplied) S. coelicolor spore stocks were diluted to 2x108 cfu/mL and 0.5ul was spotted onto a 60x15mm petri dish with 4ml ISP2 agar, resulting in agar approximately 2 mm thick. For interactions, 0.5 ul of an Amycolatopsis sp. AA4 stock (8x108 cfu/mL) was spotted 0.75 cm away from the S. coelicolor spot. Interactions were grown for 4 days at 30°C, then imaged or harvested for RNA isolation. Biomass was collected after 4 days of growth at 30°C using a cell scraper. more...
