Amino acids availability and subsequent metabolic reprogramming sustain endocrine resistance in breast cancer (microRNA)

(Submitter supplied) The majority of breast tumours express oestrogen receptor (ER) and receive endocrine therapy (ET) as standard care. Despite its efficacy, ~40% of women relapse with endocrine therapy-resistant (ETR) disease. A global transcription analysis and subsequent integrative approaches of ETR cells showed a deregulated node between miR-23b-3p and the amino acids transporter SLC6A14. Mechanistic analysis supports a model in which such deregulation impairs amino acids metabolism in the ETR cells with subsequent activation of autophagy and enhanced aspartate and glutamate import mediated by the SLC1A2 transporter. more...

miRNA expression data through TNF-alpha-signaling in human umblical vein endothelial cells (HUVECs)

(Submitter supplied) Lysine 9 di-methylation and lysine 27 tri-methylation of histone H3 (H3K9me2 and H3K27me3) are mostly linked to gene repression. However, functions of repressive histone methylation dynamics during inflammatory responses remain poorly understood. Here, we show that lysine demethylase 7A (KDM7A) and 6A (UTX) are rapidly transported to nuclear factor kappa-B (NF-κB) related elements in human endothelial cells in response to tumor necrosis factor (TNF)-α. more...

miRNA expression signatures in chronic hepatitis B (CHB) treated with nucleos(t)ide analogues (NA)

(Submitter supplied) Hepatitis B virus (HBV) infection is one of the major causes of hepatocellular carcinoma (HCC). Nucleos(t)ide analogue (NA) therapy is effective to reduce the HCC incidence, while it doesn’t completely prevent HCC. In this study, we explored the involvement of microRNA (miRNA) in the post-NA treatment HCC development. Chronic hepatitis B (CHB) patients who received NA treatment (n = 18) were divided into 2 groups: those who didn’t develop HCC during the follow-up period (non-HCC group) and those who developed HCC after treatment (HCC group). more...

Dysregulated mRNA and miRNA expression networks in Helicobacter pylori infected gastric epithelial cell line, AGS (miRNA)

(Submitter supplied) We performed genome-wide mRNA and miRNA expression analysis in gastric epithelial cell line AGS infected with Helicobacter pylori at 100 Multiplicity of Infection (MOI), comparing the expression profile of infected cells with uninfected cells. Differentially expressed mRNAs and miRNAs were identified, which showed a 1.5 fold change in expression in infected cells compared to uninfected cells. We performed in silico analysis of miRNA-mRNA target interactions among the inversely correlated differentially expressed miRNAs and mRNAs, and performed functional enrichment analysis of the target genes, to identify important cellular pathways that reflect the patho-biology of H. more...

Ionizing radiation induced changes of miRNA expression in A549 lung cancer cells

(Submitter supplied) We have employed microarray expression profiling as a discovery platform to identify microRNAs induced by radiation. Human A549 lung cancer cells were irradiated (2Gy/day for consistent 3days) and miRNA signature was identified that distinguished between control and radiation treated samples.Irradiated and un-irradiated A549 cells were harvested after 48 hr of treatment and microarray analysis was performed. more...

Characterizing the global changes in miRNA expression in human atrial appendages with persistent atrial fibrillation.

(Submitter supplied) Atrial fibrillation (AF), the most common cardiac rhythm disorder, is a major cause of cardiovascular morbidity and mortality. AF is characterized by the rapid and irregular activation of the atrium with diverse abnormalities, including electrical, structural, metabolic, neurohormonal, or molecular alterations.3 Although the pathophysiology of AF is complex, it has traditionally been treated with antiarrhythmic drugs that control the rhythm by altering cardiac electrical properties, principally by modulating ion channel function. more...

Comparison of gene expression profiles between myxoid and round cell component in myxoid liposarcoma

(Submitter supplied) Round cell component is a resk factor associated with metastasis and poor prognosis in myxoid liposarcoma. To identify microRNAs which play a role in the malignancy of round cell component, we performed microRNA microarray analysis and compared the miRNA expression profilings between myxoid and round cell component.

Identification of microRNA profile specific to Cancer Stem-Like Cells directly isolated from Human Larynx Cancer Specimens (Agilent-031181)

(Submitter supplied) Our results showed that certain miRNAs in CD133high cells could be used as cancer stem cell markers. Based on these results, we propose that this panel of miRNAs might carry crucial roles in LCa initiation, progression, metastasis, chemoresistance and recurrence through regulating stem cell properties of tumor cells.

microRNA expression signatures for colon cancer initiating cell in CRC

(Submitter supplied) In CCICs, G9a expression significantly increases K-RAS protein expression but has no effect on mRNA level. This raises the question of whether microRNA (miRNA) is involved in regulation. A heat map of a miRNA array in G9a versus shG9a transfectants ranked the Let-7 family first, with only Let-7b consistently up-regulated in G9a-knockdown CCICs and down-regulated in G9a-overexpressed non-CCICs

miRNAs as Common Regulators of the Transforming Growth Factor (TGF)-β Pathway in the Preeclamptic Placenta and Cadmium-treated Trophoblasts: Links between the Environment, the Epigenome and Preeclampsia

(Submitter supplied) Preeclampsia (PE) is a pregnancy disorder characterized by high blood pressure and proteinuria that can cause adverse health effects in both mother and fetus. There is no current cure for PE other than delivery of the fetus. While the etiology is unknown, poor placentation of the placenta due to aberrant signaling of growth and angiogenic factors has been postulated as causal factors of PE. In addition, environmental contaminants, such as the metal cadmium (Cd), have been linked to placental toxicity and increased risk of developing PE. more...

A Novel role for GSK3B as a modulator of Drosha Microprocessor activity and microRNA biogenesis

(Submitter supplied) Regulation of microRNA (miR) biogenesis is complex and stringently controlled. Here we identify the kinase GSK3B as an important modulator of global miR biogenesis at microprocessor level. Repression of GSK3B activity reduces Drosha activity towards pri-miRs, leading to accumulation of unprocessed pri-miRs and reduction of pre-miRs and mature miRs without altering levels or cellular localisationof miR biogenesisproteins. more...

Downregulation of microRNA-132 is associated with liver metastasis of colorectal cancer by targeting Anoctamin 1.

(Submitter supplied) In this study, we conducted a microarray-based analysis to identify differentially expressed miRNAs in CRC by comparing miRNA profiles among primary CRC tissues from patients with liver metastases, primary tissues without liver metastases, and liver metastatic lesions. microRNAs (miRNAs) have been shown to have a potential for cancer diagnosis lately. The main objective of this study is to identify a novel biomarker serum miRNA from the patients with colorectal cancer (CRC). more...
