Tumor grafts derived from patients with head and neck squamous carcinoma.

(Submitter supplied) Head and neck squamous cell carcinoma (HNSCC) is a deadly and disfiguring disease for which better systemic therapy is desperately needed. The development of new therapies for HNSCC and the understanding of its biology both depend upon clinically relevant animal models. An increasingly promising xenograft model, the patient derived xenograft (PDX), is developed by surgically implanting tumor tissue directly from a patient into an immunocompromised mouse. more...

Patient-derived xenograft models of non-small cell lung cancer for evaluating targeted drug sensitivity and resistance

(Submitter supplied) Background: Patient-derived xenograft (PDX) models are a useful tool in cancer biology research. However, the number of lung cancer PDXs is limited Results: In the present study, we successfully established ten PDXs, including three adenocarcinoma (AD), six squamous cell carcinoma (SQ) and one large cell carcinoma (LA), from 30 patients with non-small cell lung cancer (NSCLC) (18 AD, 10 SQ, and 2 LA), mainly in SHO mice (Crlj:SHO-PrkdcscidHrhr). more...

Over-expression of BAG-1 in head and neck squamous cell carcinomas (HNSCC) is associated with cisplatin-resistance

(Submitter supplied) In order to improve therapy for head and neck squamous cell carcinoma (HNSCC), biomarkers associated with local and/or distant tumor relapses and cancer drug resistance are urgently needed. This study identified a potential biomarker, BAG-1 (Bcl-2 associated athanogene-1), that is implicated in HNSCC insensitive to cisplatin and tumor progression. Advanced HNSCC cells revealed resistant to cisplatin accompanied by increased expression of BAG-1 protein. more...

Gene expression data from head and neck cancer patient derived xenografts

(Submitter supplied) Head and neck cancer is a hetergeneous disease. Based on previoulsy defined molecular subtypes we associated gene expression with response to different compounds. We used microarry gene expression for molecular subtyping Cetuximab is the single targeted therapy approved for the treatment of head and neck cancer (SCCHN). Predictive biomarkers have not been established in the clinical setting and patient stratification based on molecular tumor profiles has not been possible. more...

The dual pathway inhibitor rigosertib is effective in direct-patient tumor xenografts of head and neck squamous cell carcinomas

(Submitter supplied) Rigosertib treatment of head and neck squamous cell cancer The dual pathway inhibitor rigosertib inhibits phosphoinositide 3-kinase (PI3K) pathway activation as well as polo-like kinase 1 (PLK1) activity across a broad spectrum of cancer cell lines. The importance of PIK3CA alterations in head and neck squamous cell cancer (HNSCC) has raised interest in exploring agents targeting PI3K, the product of PIK3CA. more...

Differential RNA expression in human neck and squamous cell carcinoma cell lines (T-HEp3 and D-HEp3).

(Submitter supplied) We report the RNA expression profile in the tumorigenic (T-HEp3) and dormant (D-HEp3) HNSCC cell lines. T-HEp3 is a human epidermoid carcinoma, which grows on chorioallantoic membreane and metastasizes with high efficency into the organs of chick embryo. Upon serial paasage for more than 50 times in vitro , T-HEp3 loses tumorigenic and metastatic potential and termed as D-HEp3. This is an ideal cell line model system to study cancer dormancy.

Transcriptional dissection of pancreatic tumors engrafted in mice

(Submitter supplied) Engraftment of primary pancreas ductal adenocarcinomas (PDAC) in mice to generate patient derived xenograft (PDX) models is a promising platform to for biological and therapeutic studies in this disease. However, these models are still incompletely characterized. Here, we measured the impact of the murine environment on the gene expression of the engrafted human tumoral cells. We have analyzed gene expression profiles from 35 new PDX models and compared them with previously published microarray data from PDAC and hepatocellular carcinoma (HCC). more...

Gene expression profiles of a non-gestational choriocarcinoma and its corresponding patient-derived xenograft tumor

(Submitter supplied) Non-gestational choriocarcinoma (NGC) is a rare subtype of malignant germ cell tumor. We established a patient-derived xenograft (PDX) model using tumor tissue from an 18-year-old patient with NGC. Then, we performed mRNA-seq analysis using the fresh-frozen tissues of the original tumor and the PDX model.

Improved patient-derived tumor models in pancreatic ductal adenocarcinoma employing orthotopic implantation

(Submitter supplied) Pancreatic ductal adenocarcinoma has a very poor prognosis, and new therapies and preclinical models are urgently needed. We developed patient-derived xenografts (PDXs), established PDX-derived cell lines (PDCLs), and generated cell line-derived xenografts (CDXs), and integrated these to create 13 matched trios, as systematic models for this cancer. Orthotopic implantation (OI) of PDCLs showed tumorigenesis and metastases to the liver and peritoneum. more...
