Effects of FGF2 and cell culture surface on the transcriptome of adult human dermal fibroblasts

(Submitter supplied) The current study set to determine the effects of Fibroblast Growth Factor 2 and cell culture surface (tissue culture plastic and glass) on the transcriptome of adult human dermal fibroblasts. Transcriptional profiles of adult human dermal fibroblasts grown in four experimental conditions (glass, glass and addition of FGF2, plastic, plastic and addition of FGF2) were compared. Comparison of the transcriptomes will allow to identify significantly differentially expressed genes due to FGF2 and surface treatment, which in turn will allow for identification of the pathways affected by these factors in the human adult fibroblasts.

(Submitter supplied) The intracellular pathogen Trypanosoma cruzi secretes an activity that blocks TGF-β-dependent induction of connective tissue growth factor (CTGF/CCN2). Transforming growth factor-β effect on fibroblasts in the presence of Trypanosoma cruzi conditioned medium

Analysis of fibroblasts stimulated with TGF-β in the presence of T. cruzi-conditioned medium (PCM). A soluble factor(s) from T. cruzi inhibits TGF-ß-induced MAP kinase activation. Results provide insight into the ability of secreted parasite molecules to hinder agonist-induced host cell signaling.

Fibroblast Growth Factor-2 bound to specific dermal fibroblasts-derived extracellular vesicles is protected from degradation: relevance to wound healing

(Submitter supplied) Fibroblast growth factor-2 (FGF2) has multiples roles in cutaneous wound healing but its natural low stability prevents the development of its use in therapeutic for impaired skin repair. Here we show that FGF2 binds the outer surface of dermal fibroblast (DF)-derived extracellular vesicles (EVs) and this association protects FGF2 from fast degradation. We performed RNA-seq analysis after EV treatment to examine modulated genes in DFs.

Genome-wide analysis of gene expression in response to leptin and IL-1β in gingival fibroblasts

The hypothesis tested in the present study was that leptin and IL-1β synergistically effect the phenotype of gingival fibroblasts. Effects of 1,25-dihydroxyvitamin D (1,25D) on global gene expression of human fibroblasts with (CO) or without (MUT) a functional VDR

(Submitter supplied) Human skin fibroblasts from an individual with hereditary vitamin D-resistant rickets bearing a homozygous p.Arg30* VDR mutation [Damiani et al., Osteoporos Int 2015; 26(6):1819-23] and from an age/sex-matched control were obtained from 4-mm punch biopsies of the forearm skin, after institutional board approval and with informed consent.
