Transcriptome of the demosponge Amphimedon queenslandica at life cycle transitions

(Submitter supplied) Poriferans, like the demosponge Amphimedon queenslandica, are the earliest diverging metazoans and may hold the key to understanding the evolution of metazoan pathways. A. queenslandica exhibits a biphasic lifecycle, with free-swimming larvae and sessile adult stages. To study changes in the transcriptome during this period of morphological and ecological transition, we generated poly(A) fragment libraries for A. more...

Stem cell differentiation trajectories in Hydra resolved at single cell resolution

(Submitter supplied) The adult Hydra polyp continuously renews all of its cells using three separate stem cell populations but the genetic pathways enabling these dynamic tissue properties are not well understood. We used Drop-seq to sequence 24,985 Hydra cells and identified the molecular signatures of a broad spectrum of cell states, from stem cells to terminally differentiated cells. We constructed differentiation trajectories for each cell lineage and identified the transcription factors expressed along these trajectories, thus creating a comprehensive map of all developmental lineages in the adult animal. more...

Ephydatia muelleri methylome

(Submitter supplied) Ephydatia muelleri is a cosmopolitan freshwater demosponge, with potential to become a model system. We have participated in a large collaborative project to sequence the genome (PRJNA579531), methylome, transcriptome for this species, aiming to better understand the biology of this sponge species. In terms of DNA methylation, it presents relatively low methylation levels compared to the methylomes of other sponges (A. more...

Time-driven molding of the epidermal stem cell transcriptome

(Submitter supplied) Epidermal stem cells ensure proper faring of skin homeostatic processes under both physiological and challenging conditions. Currently, the molecular events underpinning ageing within the epidermal stem cell niche are poorly understood. Here, we report that epidermal stem cells are endowed with a collection of time-sensitive genes which constitute specific functional programs that become deregulated in an age-dependent manner.
