RNA-sequencing of CD4+ T cells from mice prior to and after infection with influenza virus, and that were exposed developmentally to vehicle or TCDD

(Submitter supplied) We used next generation RNAsequencing to identify how developmental exposure to TCDD alters gene expression in CD4+ T cells. Specifically, mice were developmentally exposed to vehicle or TCDD. At adulthood, CD4+ T cells were purified from peripheral lymph nodes of naive animals or the mediastinal lymph nodes of influenza A virus (IAV) infected mice (9 days post infection). RNA was isolated from these cells, and RNA-seq was performed. more...

Whole Genome Bisulfite Sequencing of CD4+ T cells from mice developmentally exposed to vehicle or TCDD prior to and during influenza infection

(Submitter supplied) We used Whole Genome Bisulfite Sequencing (WGBS) to compare DNA methylation patterns across the genome of CD4+ T cells. Specifically, mice were developmentally exposed to vehicle or TCDD. At adulthood, CD4+ T cells were purified from peripheral lymph nodes of naive animals or the mediastinal lymph nodes of influenza A virus (IAV) infected mice (9 days post infection). RNA sequencing of CD4+ T cells from mice developmentally exposed to TCDD

(Submitter supplied) We used next generation RNAsequencing to identify how triggering the aryl hydrocarbon receptor (AHR) during development alters T cell gene expression. Specifically, mice were developmentally exposed to vehicle or the AHR agonist TCDD. At adulthood, mice were infected with influenza A virus. CD44hiCD4+ and CD44loCD4+ T cells were FACs purified from the mediastinal lymph nodes on day 9 post infection. more...

Linking the aryl hydrocarbon receptor with altered DNA methylation patterns and developmentally induced aberrant antiviral T cell responses [RNA-Seq]

(Submitter supplied) Successfully fighting infection requires a properly tuned immune system. Recent epidemiological studies link exposure to pollutants that bind the aryl hydrocarbon receptor (AHR) during development with poorer immune responses later in life. Yet, how developmental triggering of AHR durably alters immune cell function remains unknown. Using a mouse model, we show that developmental activation of AHR leads to long-lasting reduction in the response of CD8+ T cells during influenza virus infection, cells critical for resolving primary infection. more...

Postnatal administration of S-adenosylmethionine restores developmental AHR activation-induced deficits in CD8+ T cell function during influenza A virus infection

(Submitter supplied) Developmental exposures can influence life-long health; yet, approaches to counteract negative consequences of developmental toxicity are limited due to poor understanding of cellular mechanisms. The aryl hydrocarbon receptor (AHR) binds many small molecules, including numerous pollutants. Developmental exposure to the signature environmental AHR ligand 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) significantly dampens the adaptive immune response to influenza A virus (IAV) in adult offspring. more...

Dendritic cell response to 2,3,7,8-tetrachlorodibenzo-p-dioxin-induced aryl hydrocarbon receptor activation

Analysis of MACS-sorted splenic dendritic cells (DC) from aryl hydrocarbon receptor (AHR) +/+, C57BL6 females after 24hr in vivo treatment with TCDD. The TCDD ligand is an AHR agonist and a potent immunotoxicant. Results provide insight into the consequences of TCDD-induced AHR activation in DCs.

AHR protects against lung vascular leakage in viral infection

(Submitter supplied) We have identified the apelin-APJ axis as a mechanism of protection mediated by AHR signalling in lund=g endothelial cells. This RNAseq experiment will compare the effects of apelin receptor blockade on endothelial and epithelial responses in the lung during influenza infection.

Aryl hydrocarbon receptor (AHR)-dependent protection against lung vascular leakage in viral infection

(Submitter supplied) Disruption of the lung endothelial-epithelial cell barrier during respiratory virus infection causes cell and fluid accumulation in the air spaces and compromises vital gas exchange function. Endothelial cell dysfunction is known to exacerbate tissue damage; however, it is unclear whether the lung endothelium engages in tissue-protective activity during viral infection. Here we show that the environmental sensor aryl hydrocarbon receptor (AHR) is predominantly active in endothelial cells and protects against influenza-induced lung damage. more...
