Cancer stem cell population in highly metastatic variants of MDA-MB-231 breast cancer cell line

Analysis of CD24-/CD44+/ESA+ cancer stem cells from MDA-MB-231 breast cancer (BC) cell line and its highly metastatic variants 231BoM-1833 and 231BrM-2a. The majority of BC deaths are attributed to metastatic disease. Results provide insight into the molecular basis of metastatic progression.

Expression data from CD44-knockdown MDA-MB-231 human breast cancer cells.

(Submitter supplied) CD44, an adhesion molecule that binds to extracellular matrix, primarily to hyaluronan (HA), has been implicated in cancer cell migration, invasion, and metastasis. CD44 has also recently been recognized as a marker for stem cells of several types of cancer. However, the roles of CD44 in the development of bone metastasis still remain unclear. To explore this issue, we established the MDA-MB-231 human breast cancer cells stably expressing short hairpin RNA against CD44.

Gene expression of S100A10 knocked down human breast cancer cells

(Submitter supplied) The expression level of S100A10 was much higher in the CD44+ cancer cells metastasized to the liver than those at the primary site in the breast cancer patient-derived tumor xenograft mice. Expression level of S100A10 increased by the oncoprotein, PML/RARα and RAS, and its higher expression is linked to worse outcome in a number of cancer types including breast cancers

Gene expression of breast cancer stem cells.

(Submitter supplied) Metastatic progression remains the major cause of death in human breast cancer. Cancer cells with cancer stem cell (CSC) properties drive initiation and growth of metastases at distant sites. We have previously established the breast cancer patient-derived tumor xenograft (PDX) mouse model in which CSC marker CD44+ cancer cells formed spontaneous microscopic metastases in the liver. In this PDX mouse, the expression levels of S100A10 and its family proteins were much higher in the CD44+ cancer cells metastasized to the liver than those at the primary site.

Effect of endothelial-specific endorepellin expression on gene expression of breast cancer allografts in Tie2CreER;ERKi mice

(Submitter supplied) To explore the translational implications of endorepellin in breast cancer, we created a double transgenic, inducible Tie2CreER;ERKi mouse line that expresses recombinant endorepellin specifically from the endothelium. We investigated the therapeutic effects of recombinant endorepellin overexpression in an orthotopic, syngeneic breast cancer allograft mouse model. Tamoxifen-induced expression of recombinant endorepellin specifically from the endothelium in Tie2CreER;ERKi mice markedly suppressed breast cancer allograft growth, hyaluronan deposition in the tumor proper and perivascular tissues, and tumor angiogenesis.

Hyaluronan-CD44 signaling promotes pro-tumor inflammation in breast cancer
