miR-182, miR-221 and miR-222 are potential urinary extracellular vesicle biomarkers for canine urothelial carcinoma

(Submitter supplied) Urothelial carcinoma (UC) is the most common tumour type in canine bladder cancer. Current diagnostic methods are technically challenging or can lack specificity, hence there is a need for novel biomarkers of UC. To this end, we analysed the microRNA (miRNA) cargo of extracellular vesicles (EVs) from urine samples of dogs with UC to identify miRNAs that could be utilised as biomarkers. Urine was fractionated using ultrafiltration combined with size-exclusion chromatography and small RNA sequencing analysis was performed on both the EV enriched and (EV free) protein fractions. more...

Host-Microbe Interactions in the Nasal Cavity of Dogs with Chronic Idiopathic Rhinitis

(Submitter supplied) Chronic rhinitis (CR) is a frustrating clinical syndrome in dogs and our understanding of the disease pathogenesis in is limited. Increasingly, host-microbe interactions are considered key drivers of clinical disease in sites of persistent mucosal inflammation such as the nasal and oral cavities. Therefore, we applied next generation sequencing tools to interrogate abnormalities present in the nose of dogs with CR and compared immune and microbiome profiles to those of healthy dogs. more...

Analysis of gene expression in the dog macular retina and supporting tissues: comparison with human macular gene expression profiles

(Submitter supplied) Dogs are commonly used models of human inherited retinal disorders. Because the dog retina contains a macula-like region, dogs are susceptible to maculopathies with many shared genetic etiologies with humans. Human macular gene expression has been characterized and provides insight into the underlying basis of macular disease. We sought to compare macular gene expression profiles in dogs and humans and interrogate macular disease-associated genes for differential expression between macula and periphery. more...

Deconstruction of cellular dynamics after treatment of volumetric muscle loss injury with extracellular matrix

(Submitter supplied) Volumetric muscle loss (VML) is an acute loss of a critical volume of skeletal muscle that results in inflammation, fibrotic scarring in muscle tissue, and permanent muscle defects. The cellular and molecular processes that underlie fibrosis induced from VML injury need to be evaluated in larger animal models. Therefore, we used a canine model of VML alongisde treatment with an extracellular matrix (ECM) hydrogel. more...

Stromal expression profiling reveals immune-driven adaption to malignancy in canine melanoma subtypes

(Submitter supplied) Canine mucosal melanoma (CMM) is the most common oral malignancy in dogs and exhibits significantly higher aggressiveness than its cutaneous counterpart (CCM), yet the reasons for this disparity remain unclear. The influence of cancer-associated stroma (CAS) on tumour progression is well-recognized, but a detailed understanding of CAS in canine melanoma is missing. To comprehensively assess stromal reprogramming in canine melanoma, we analysed CAS from 21 CMM, 14 CCM and normal stroma from 10 skin and 9 oral mucosa samples by laser-capture microdissection followed by RNA sequencing. more...

Spatial gene expression of canine veins during carotid-cartoid vein bypass implantation

(Submitter supplied) Vein graft failure (VGF) following cardiovascular bypass surgery results in significant patient morbidity and cost to the healthcare system. Vein graft injury can occur during autogenous vein harvest and preparation, as well as after implantation into the arterial system, leading to the development of intimal hyperplasia, vein graft stenosis, and, ultimately, bypass graft failure. While previous studies have identified maladaptive pathways that occur shortly after implantation, the specific signaling pathways that occur during vein graft preparation are not well defined and may result in a cumulative impact on VGF. more...

Establishment and characterization of six canine hepatocellular carcinoma cell lines (Transcriptional changes induced by drug treatment with toceranib or sorafenib)

(Submitter supplied) Hepatocellular carcinoma (HCC) is the most common malignant liver tumor in dogs. Although surgical resection is a major treatment option for canine HCC, there are no distinct strategies for unresectable tumor subtypes or adjuvant chemotherapy for tumors with positive margins. We aimed to establish and characterize novel HCC cell lines from canine patients. The cellular morphology, general growth features and tumorigenicity of the established cell lines were evaluated. more...

Canid alphaherpesvirus 1 infection affects the gene expression and secretome profile of canine adipose-derived mesenchymal stem cells in vitro

(Submitter supplied) Canine adipose -derived mesenchymal stem cells (cAD-MSCs) demonstrate promising tissue repair and regeneration capabilities. However, the procurement and preservation of these cells or their secreted factors for therapeutic applications pose a risk of viral contamination, and the consequences on cAD-MSCs remain unexplored. Consequently, this research sought to assess the impact of Canid alphaherpesvirus 1 on the functional attributes of cAD-MSCs, including gene expression profiles and secretome composition. more...

Altered Chromatin State Drives Hypersocial Behaviors in Canids

(Submitter supplied) Polymorphic transposable elements found in regulatory sequences can impact the associated transactional modules. We investigated such molecular alterations for these elements in the dog genome that evolved from directional selection during domestication. We find that the polymorphic element in canine GTF2I intron 17 is associated with altered looping, and hence distinct cis-regulatory landscapes. We report supporting evidence of an E2F1-DNA binding peak concordant with the altered loop and higher expression of GTF2I exon 18. more...

An optimized workflow for transcriptomic analysis from archival paraformaldehyde-fixed retinal tissues collected by laser capture microdissection

(Submitter supplied) RNA sequencing (RNA-seq) coupled with laser capture microdissection (LCM) is a powerful tool for transcriptomic analysis in unfixed fresh-frozen tissues. Fixation of ocular tissues for immunohistochemistry commonly involves the use of paraformaldehyde (PFA) followed by embedding in Optimal Cutting Temperature (OCT) medium for long-term cryopreservation. However, the quality of RNA derived from such archival PFA-fixed/OCT-embedded samples is often compromised, limiting its suitability for transcriptomic studies. more...

Extracellular vesicles from epicardial fat mediate long-distance heart-brain communication and promote atrial fibrillation associated cognitive impairment (EAT exosomal miRNA-seq and hippocampal mRNA-seq)

(Submitter supplied) 2 weeks of rapid atrial pacing increased the release of epicardial adipose tissue (EAT) exosomes. The red fluorescence of Ad-CD63-RFP traces the increase of EAT exosomes in the canine hippocampus. RNA Sequencing showed that the differentially expressed genes of cfa-miR-22e in EAT exosomes were significantly increased, and its target gene of IL33 was down-regulated in the hippocampus. Exosomes secreted by EAT in AF model canine penetrate the BBB, and activate hippocampal microglia through cfa-miR-22e/IL33 signaling pathway, which may be related to the increased risk of cognitive impairment in AF.
