Compartmentalisation proteomics revealed endolysosomal protein network changes in goat model of atrial fibrillation

(Submitter supplied) To identify the role of the endolysosomal (EL) protein network in atrial fibrillation (AF), we applied a newly developed organelle isolation method to the AF goat model. To accomplish the goal, we used proteomics, transcriptomics, integrated analysis, enzyme assays, electron tomography, and western blotting and identified several regulating EL proteins, related protein networks and pathways.

Transcriptome analysis revealed the characteristics and functions of long non-coding RNAs in the hypothalamus during sexual maturation in goats

(Submitter supplied) To reveal the transcriptomic changes of hypothalamic tissue in goats from birth to sexual maturity, we performed transcriptome sequencing of Jining Grey Goat hypothalamic tissues at four developmental stages after birth. A total of 20 libraries from four different development stages (5 goats per stage) were successfully constructed, and the corresponding mRNA and lncRNA expression profile were obtained.

Divergent composition and transposon-silencing activity of small RNAs in mammalian oocytes

(Submitter supplied) We found piRNAs with different lengths represented the predominant small RNA species in oocytes from the 12 explored species, except mouse. We found endo-siRNAs resulted from the truncated Dicer isoform were mouse-specific, and os-piRNAs associating with PIWIL3 in human oocytes are widespread in mammals and are typically with low levels of the 2’-3’-O-methylation. The sequences of many highly expressed piRNA clusters are fast-evolving compared with their syntenic genomic locations, and the TE families distributing in the conserved piRNA clusters are various between species.

Integrating into dairy goats chromatin accessible sites during lactation promotes transgene expression efficiency

(Submitter supplied) Combining RNA-seq with ATAC-seq, we screened the open chromatin of dairy goat mammary epithelial cells during lactation and then integrated human lysozyme into various open regions using the CRISPR/Cas9 system. A comparison of lysozyme gene expression and integration efficiency showed that chromatin accessibility at target sites was significantly positively correlated with gene insertion efficiency, and the insertion of foreign genes into highly accessible chromatin regions could enhance its expression.

Milk fat miRNome changes in response to LPS challenge in Holstein cows

(Submitter supplied) Cows were fed a lactation diet at ad libitum intake (n = 6). At 27±3 days in milk, cows were injected with 50 µg of LPS E. coli in one healthy rear mammary quarter. Milk samples were collected just before LPS challenge (LPS-) and 6.5 h after LPS challenge (LPS+) from the same cows. Microarray analysis was performed using customized 8x60K ruminant miRNA microarrays to compare LPS- to LPS+ miRNome. MiRNome comparison between LPS- and LPS+ identified 37 differentially abundant miRNAs (q-value ≤ 0.05)
