Primary mesenchymal cells from the human fallopian tube infected with Chlamydia trachomatis

(Submitter supplied) We profiled metabolite changes upon C.trachomatis infection in primary human cells in order to identify rewiring of metabolite networks due to infection. In addition, gene expression micro array analyses were used to determine concurrent transcriptional regulation.

Gene expression profiles of human patient derived ectocervical stem cells and 3D organoids with and without Chlamydia trachomatis infection

(Submitter supplied) Coinfections with pathogenic microbes continually confront cervical mucosa, yet their implications in pathogenesis remain unclear. Lack of in-vitro models recapitulating cervical epithelium has been a bottleneck to study coinfections. Using patient-derived ectocervical organoids, we systematically modelled individual and coinfection dynamics of Human papillomavirus (HPV)16 E6E7 and Chlamydia, associated with carcinogenesis. more...

Gene expression signatures after immunisation with an HIV-1 recombinant Vesicular Stomatitis Virus (VSV) vaccine following three priming doses of IL-12 pDNA enhanced HIV-1 multi-antigen pDNA vaccine (BIOVACSAFE protocol HVTN087)

(Submitter supplied) The time course of whole blood gene expression was measured after the fourth immunisation at month 6 in participants who had been randomised into vaccine or placebo group 1 of the clinical protocol HVTN087 - A phase 1 trial to evaluate the safety, tolerability, and immunogenicity of an IL-12 pDNA enhanced HIV-1 multi-antigen pDNA vaccine delivered intramuscularly with electroporation, with an HIV-1 rVSV vaccine boost, in healthy HIV-uninfected adult participants (NCT01578889).

Gene expression signatures after immunisation with Boostrix - alum adsorbed tetanus toxoid, reduced diphtheria toxoid and acellular pertussis vaccine (BIOVACSAFE protocol 305D2)

(Submitter supplied) The time course of whole blood gene expression was determined in healthy adult human participants who were randomised to receive either a single intramuscular immunisation with Boostrix vaccine (characteristics: treatment = BOOSTRIX) or equivalent volume saline control (characteristics: treatment = PLACEBOD2). Participants are identified who reported having had an immunisation with Boostrix within the past 5 years (characteristics: recent boostrix = 1).

Epithelial response to IFN-γ promotes SARS-CoV-2 infection

(Submitter supplied) SARS-CoV-2, the agent causing COVID-19, invades epithelial cells, including those of the respiratory and gastrointestinal mucosa, using angiotensin-converting enzyme-2 (ACE2) as a receptor. Subsequent inflammation can promote rapid virus clearance. However, severe cases of COVID-19 are characterized by an inefficient immune response that fails to clear infection. Using primary epithelial organoids from the colon, we explored how IFN-γ, a central antiviral mediator elevated in COVID-19, affects differentiation, ACE2 expression, and infectivity with SARS-CoV-2. more...

Gene expression profiles of primary human gall bladder epithelial cells with and without infection with S.paratyphi WT and ∆cdtB mutant for 2 and 7 days

(Submitter supplied) We have successfully established organoid culture model for human and murine gall bladder (GB) epithelial cells allowing long-time culture of primary epithelial cells. This model was used to analyze factors influencing stemness and to study infection with Salmonella paratyphi and its toxin Cdtb.

Gene expression signatures in blood and muscle biopsy after intramuscular immunization of healthy adult humans with adjuvanted vaccines (BIOVACSAFE protocol 305E)

(Submitter supplied) The gene expression in whole blood and simultaneous muscle biopsy (site of immunization and control contralateral non-injected leg) was measured in healthy adult male humans attending a research unit (CRC, University of Surrey, UK), who were randomized to receive a single immunization with an adjuvanted licensed vaccine or saline placebo control.

Gene expression profiles of human cervical primary cells from ecto- and endocervix

(Submitter supplied) The cellular origin of cervical cancers remains unclear. Revealing molecular details of transformation in this tissue has been hampered by the lack of culture systems, resembling the in vivo cervical architecture. Here we established a long-term in vitro 3D cervical organoid model derived from stem cells of human or mouse cervical tissue which recapitulates the in vivo stratified ectocervical and columnar endocervical epithelium. more...

Chronic Chlamydia infection in fallopian tube organoids alters epithelial differentiation and accelerate methylation changes

(Submitter supplied) Chlamydia trachomatis (Ctr) is able to colonize the human fallopian tube, causing scarring and inflammation, which can lead to infertility. Here we describe a chronic Ctr infection model using human fallopian tube organoids. The defense response of the organoids to infection includes active expulsion of the bacteria from the apical side of the epithelium and compensatory cellular proliferation, clearly demonstrating a role for core mechanisms of epithelial homeostasis in defense against this invading pathogen. more...

Gene expression profile of human lung epithelial cells with different culture methods.

(Submitter supplied) The goal of our study is to establish a method to propagate human pneumocytes efficiently in a large scale. While human pneumectomy-derived lung epithelial cells (LECs) showed efficient long-term growth when co-cultured with irradiated NIH-3T3, they do not express molecular markers of mature pneumocytes or airway cells in this condition. Removal of feeders causes differentiation toward airway club cell-like phenotype, and an induction treatment with combination of small molecules and growth factors is required to differentiate them toward pneumocytes. more...
