Transcriptional response of FZF1 alleles from S. cerevisiae and S. paradoxus to the addition of sulfite

(Submitter supplied) Transcriptional profiling of four different yeast FZF1 alleles: S. cerevisiae, S. paradoxus and two reciprocal chimeras with the coding and 5' noncoding region from opposite species of S. cerevisiae and S. paradoxus, both before and 15 minutes after sulfite addition. FZF1 is a transcription factor that is known to be turned on in response to sulfite. Here we determine whether the FZF1 allele from two species leads to different transcriptional responses and the effect of the individual noncoding and coding regions on the transcriptional response.

PAT-seq: a simple approach to digital gene expression, the measure of poly(A)-tail length and its position in eukaryotic transcriptomes

(Submitter supplied) The Poly(A)-Tail focused RNA-seq, or PAT-seq approach, is an affordable and efficient tool for the measure of 3’UTR dynamics. We show here that PAT-seq returns (i) digital gene expression, (ii) polyadenylation site usage within and between samples, including alternative adenylation, and (iii) the polyadenylation-state the transcriptome. PAT-seq differs from previous 3’ focused RNA-seq methods in that it strictly depends on native 3’ adenylation within total RNA samples and thus removes the need for ribosome depletion and, that the full native poly(A)-tail is included in the sequencing libraries. more...

Comparative transcriptomic analysis between a high and a low sulfite producers wine yeast strains

The Msn2/4-like gene transcription factor Com2 (ORF YER130) is a key player of S. cerevisiae response and tolerance to sulphur dioxide

(Submitter supplied) Sulphur dioxide (SO2) and its derivatives are widely used as preservatives in food and beverage industries to assure the maintenance of microbiological quality of their products. In this work it is shown, for the first time, the essential role played by the transcriptional regulator Com (ORF YER130c) in S. cerevisiae response and tolerance to SO2, this being the first biological function attributed for this regulator.
