Daily Patterns of Retinal and Choroidal Gene Expression and Circadian Biology in Myopia Pathogenesis

(Submitter supplied) We investigated diurnal changes in gene expression in the retina and choroid at the onset of experimental myopia. We induced visual form deprivation myopia in young chicks, and isolated retinal and choroidal tissues separately in form- deprived and contralateral control eyes every 4 hours over a single 24-hour period.

Retinal gene expression in chicks during imposed myopic defocus

(Submitter supplied) The retina plays an important regulatory role in ocular growth. To screen for new retinal candidate genes that could be involved in the inhibition of ocular growth, we used chick microarrays to analyze the changes in retinal mRNA expression after myopic defocus was imposed by positive lens-wear. Chicks were raised under a 12h-light/12h-dark cycle (light-onset: 8:00 am and light-offset: 8:00 pm) with unrestricted access to water and food. more...

Gene expression across 4 and 24 hours of growth modulating paradigms in chicks

(Submitter supplied) Using RNA-Seq, mRNA from 54 retinae were sequenced from male chicks in which myopia development was inhibited by one of five growth inhibitory treatments.

Chick myopia

(Submitter supplied) One-day old white Leghorn chicks were housed in brooders with a 12 hr light:dark cycle, using General Electric chroma 50 fluorescent lighting with irradiance of approximately 50μW/cm2 at chick eye level. They received Purina Chick Chow food and water ad libitum. At one week of age and at the onset of the light phase, the chicks were anesthetized with inhalation ether, and a unilateral translucent white plastic goggle was glued to the periorbital feathers to induce ipsilateral form-deprivation myopia, alternating between the left or right eye. more...

Image defocus and altered retinal gene expression in chicks

(Submitter supplied) Because refractive development is governed largely by the retina, we analyzed the retinal transcriptome in chicks wearing a spectacle lens, a well-established means to induce refractive errors, to identify gene expression alterations and to develop novel mechanistic hypotheses about refractive development.

Image defocus effect on retina/retinal pigment epithelium: time course

Analysis of retina and retinal pigment epithelium (RPE) from 1 week-old chicks wearing a unilateral spectacle lens of + or 15 diopters for 6 hours or 3 days. The spectacle lens induces refractive errors. Results provide insight into molecular mechanisms underlying refractive development.

Large-scale microRNA expression profiling identifies retinal miRNA-mRNA signaling pathways underlying form-deprivation myopia in mice

(Submitter supplied) Development of myopia is associated with large-scale changes in ocular tissue gene expression. Although differential expression of coding genes underlying development of myopia has been a subject of intense investigation, the role of non-coding genes such as microRNAs in the development of myopia is largely unknown. In this study, we explored myopia-associated miRNA expression profiles in the retina and sclera of C57Bl/6J mice with experimentally induced myopia using microarray technology. more...
