Identifying genetic determinants of Streptococcus pyogenes-host interactions in a murine intact skin infection model

(Submitter supplied) Streptococcus pyogenes is an obligate human pathobiont associated with many disease states. Here, we present a novel model of S. pyogenes infection using intact murine epithelium. From this model, we were able to perform RNA sequencing to evaluate the genetic changes undertaken by both the bacterium and host at 5 and 24 hours post infection. Analysis of these genomic data demonstrate that S. pyogenes undergoes significant genetic adaptation to successfully infect the murine epithelium, including changes to metabolism and activation of the Rgg2/Rgg3 quorum sensing (QS) system. more...

Transcriptome study of the covS-regulation of a Group A Streptococcus pyogenes M23ND

(Submitter supplied) We investigated the covS-regulation of S. pyogenes in a hypervirulent M23 strain using RNA-sequencing. The differential gene expression comparison between the covS- mutant and isogenic wild type covS+ identified altered expression of 349 (18%) genes, including a broad spectrum of virulence genes and diverse metabolic genes. The data showed that the strain achieved hypervirulence by enhancing the expression of genes responsible for invasiveness and antiphagocytosis (i.e., hasABC), by abrogating the expression of toxic genes (i.e., speB), and by compromising gene products with dispensable functions (i.e., sfb1). more...

Using RNA Seq to define the regulon of the Pseudomonas aeruginosa transcription factor Anr in low-oxygen colony biofilms.

(Submitter supplied) Purpose : The goal of this study was to use RNA Seq to define the regulon of the transciption factor Anr by comparing global transcriptional profiles of Pseudomonas aeruginosa strain PAO1 and a clinical isolate with their isogenic ∆anr mutants, grown in colony biofilms at 1% oxygen. Methods : mRNA profiles were generated for laboratory strain PAO1 and for a clinical isolate J215, as well as for ∆anr derivatives of each strain, in duplicate, by deep sequencing. more...

Transcriptomic profiling of the PhzIR quorum sensing, Gac- and PtrA- regulons of Pseudomonas chlororaphis PA23

(Submitter supplied) The biocontrol agent Pseudomonas chlororaphis PA23 protects canola (Brassica napus) against infection by the necrotrophic fungus Sclerotinia sclerotiorum. Production of PA23 secondary metabolites is governed by a complex regulatory pathway that includes the GacA/GacS two component system, the PhzI/PhzR quorum-sensing system, and a novel LysR-type transcriptional regulator, called PtrA. Through RNA-sequencing, transcriptomic profiles of PA23-WT, two quorum sensing-deficient strains, PA23-AHL and PA23-phzR, and regulatory mutants PA23-gacA, PA23-gacS and PA23-ptrA were generated allowing elucidation of the PhzRI, Gac and PtrA regulons of P. more...

Transcriptomic analysis of Streptococcus pyogenescolonizing the vaginal mucosa identifies hupY, an MtsR-regulated adhesin involved in heme utilization

(Submitter supplied) Streptococcus pyogenes (Group A Strep, GAS) is a serious human pathogen with the ability to colonize mucosal surfaces such as the nasopharynx and vaginal tract, often leading to infections such as pharyngitis and vulvovaginitis. We present genome-wide RNASeq data showing the transcriptomic changes GAS undergoes during vaginal colonization. These data reveal that the regulon controlled by MtsR, a master metal regulator, is activated during vaginal colonization. more...

Genetic and transcriptomic characteristics of RhlR-dependent quorum sensing in cystic fibrosis isolates of Pseudomonas aeruginosa: isolate E167

(Submitter supplied) In people with the genetic disease cystic fibrosis (CF), bacterial infections involving the opportunistic pathogen Pseudomonas aeruginosa are a significant cause of morbidity and mortality. P. aeruginosa uses a cell-cell signaling mechanism called quorum sensing (QS) to regulate many virulence functions. One type of QS consists of acyl-homoserine lactone (AHL) signals produced by LuxI-type signal synthases, which bind a cognate LuxR-type transcription factor. more...
