Pseudo-resistant Bacillus cereus uses biofilm-related mechanism to mimic vancomycin resistance during agar diffusion susceptibility testing

(Submitter supplied) The glycopeptide vancomycin is a drug of choice for the treatment of severe non-gastrointestinal infections with members of Bacillus cereus sensu lato. Recently, sporadic detection of vancomycin resistant phenotypes emerged, mostly for agar diffusion testing. The food packaging isolate BC70 displayed a pseudo-resistant phenotype for vancomycin in both Etest and disk diffusion. In this work, we used RNA-Seq on the nanopore platform to study differentially expressed genes in BC70 cells, which were able to actively move into the inhibition zone during vancomycin susceptibility testing using Etest and therefore appeared to be resistant. more...

Relative quantification of the recA gene for antibiotic susceptibility testing in response to ciprofloxacin for pathogens of concern

(Submitter supplied) Antibiotic resistance (AR) is one of the greatest threats to global health and is associated with higher treatment costs, longer hospital stays, and increased mortality. Current gold standard antibiotic susceptibility tests (AST) are dependent on organism growth rates resulting in prolonged diagnostic answers for slow growing organisms. Changes in the cellular transcriptome can be instantaneous in the presence of stressors such as antibiotic pressure. more...

Transcriptome analysis of Bacillus cereus strain INRA C3 in response to iron depletion and lactoferrin, accomplished by RNA sequencing

(Submitter supplied) The iron-binding glycoprotein lactoferrin exhibits anti-bacterial activity. To investigate the overall response of Bacillus cereus to lactoferrin, total transcriptome analyses were performed. Growth conditions were mMOD minimal medium, minimal medium plus 10 mg/ml lactoferrin, as well as minimal medium without iron source for comparison. Differential regulation of genes involved in iron acquisition as well as genes encoding enterotoxins and further virulence factors was of special interest.

Beyond the spore: the exosporium sugar anthrose impacts vegetative Bacillus anthracis gene regulation in cis and trans

(Submitter supplied) In preliminary experiments, the absence of anthrose in our genetic Sterne knockout was found to decrease pagA expression in BHI broth containing glucose (Fig 5A and B). This contrasted with the large spike in pagA expression when the Sterne antC mutant is grown in protein rich HIB media which does not contain sugars. When exogenous anthrose was added during growth in BHI broth, expression from PpagA increased in both WT (purple line in Fig 5A) and antC Sterne (purple line in Fig 5B). more...

SigB modulates expression of novel SigB regulon members via Bc1009 in non-stressed and heat-stressed cells revealing its alternative roles in Bacillus cereus

(Submitter supplied) Background: The Bacillus cereus Sigma B (SigB) dependent general stress response is activated via the two-component RsbKY system, which involves a phosphate transfer from RsbK to RsbY. It has been hypothesized that the Hpr-like phosphocarrier protein (Bc1009) encoded by bc1009 in the SigB gene cluster may play a role in this transfer, thereby acting as a regulator of SigB activation. Alternatively, Bc1009 may be involved in the activation of a subset of SigB regulon members. more...

Identification of Bacillus anthracis genes differentially expressed in human serum

(Submitter supplied) Bacillus anthracis causes anthrax infections in mammals. Large-scale mortality resulting from the intentional release of B. anthracis spores represents a potential bioterrorism threat. Inhalational anthrax almost invariably proceeds to fatal systemic infection, characterized by massive bacteremia. A better understanding of host-pathogen interactions is urgently needed for effective treatment of this lethal disease. more...

New genetic biomarkers to differentiate pathogenic and clinically relevant Bacillus cereus strains

(Submitter supplied) Bacillus cereus is the second leading cause of collective food poisoning in France. B. cereus is also associated with severe clinical infections leading to patient death in 10% of the cases. The emergence of B. cereus as a foodborne and opportunistic pathogen has intensified the need to distinguish strains of public health concern. In this work, by performing a screen on a large collection of B. cereus strains of varying pathogenic potential, we identified genetic determinants capable of discriminating B. more...

Direct regulons of the master virulence regulator AtxA of Bacillus anthracis

(Submitter supplied) AtxA, the master virulence regulator of Bacillus anthracis, regulates the expression of three toxins that are required for the pathogenicity of Bacillus anthracis. Recent transcriptome analyses also showed that AtxA affects a large number of genes on both chromosome and plasmid, suggesting its role as a global regulator. Its mechanism of gene regulation nor binding target in vivo was, however, not well understood. more...

Characterization of a key region involved in the NO stress response of Bacillus cereus

(Submitter supplied) Bacterial response to nitric oxide (NO) is of major importance for bacterial survival, as NO stress is a main actor of the eukaryote immune response. Several pathogenic bacteria have developed gene regulation systems involved in detoxification and in repairing the damages caused by NO. However, bacterial mechanisms of NO resistance are poorly described for Gram positive bacteria and especially Bacillus cereus. more...

Levels and Characteristics of mRNAs in Dormant Spores of Firmicute Species

(Submitter supplied) Spores of Bacillales and Clostridiales species contain 100s of different mRNAs, and their major function in Bacillus subtilis is to provide ribonucleotides for new RNA synthesis when spores germinate. In new work, RNA was isolated from spores of five Bacillales and one Clostridioides species and relative spore mRNA levels were determined by RNA-seq. Determination of RNA levels in single spores allowed calculation of RNA nt/spore, and assuming mRNA is 3% of spore RNA allowed calculation that only ~6% of spore mRNAs were present at ≥ 1/spore. more...

Plasmid-encoded Small Regulatory RNAs Regulate Chromosome Gene Expression in Bacillus anthracis

(Submitter supplied) Purpose: Small regulatory RNAs (sRNAs) are short transcripts that base-pair to mRNA targets or interact with regulatory proteins. sRNA function has been extensively studied in Gram-negative bacteria; comparatively less is known about sRNAs in Firmicutes. Here we investigate two sRNAs encoded by the virulence plasmid pXO1 of Bacillus anthracis, the causative agent of anthrax. We designated the sRNAs as as “XrrA” and “XrrB” (for pXO1-encoded regulatory RNA). more...

Regulons and protein–protein interactions of PRD-containing Bacillus anthracis virulence regulators reveal overlapping but distinct functions

(Submitter supplied) Purpose: Bacillus anthracis produces three regulators, AtxA, AcpA and AcpB, which control virulence gene transcription and belong to an emerging class of regulators termed ‘PCVRs’ (Phosphoenolpyruvate-dependent phosphotransferase regulation Domain-Containing Virulence Regulators). AtxA, named for its control of toxin gene expression, is the master virulence regulator and archetype PCVR. AcpA and AcpB are less well studied. more...
