Increased Autoreactivity and Maturity in EBI2+ Antibody-Secreting Cells from Nasal Polyps

(Submitter supplied) Rationale: Elevated numbers of antibody secreting cells (ASCs) and anti-dsDNA antibodies are found in nasal polyp (NP) tissue. Tissue anti-dsDNA IgG prospectively predicts recurrent NP, but the characteristics of the source ASCs are unknown. Objectives: To investigate whether NP B cells expressing the extrafollicular marker EBI2 have increased propensity for autoantibody production and to evaluate the molecular characteristics of NP ASCs. more...

Single-cell RNA sequencing of bladder Ewing sarcoma

(Submitter supplied) Bladder Ewing sarcoma/primitive neuroectodermal tumor (bladder ES/PNET) is a rare and highly malignant tumor associated with a poor prognosis, yet its underlying mechanisms remain poorly understood. This study employed a combination of single-cell RNA sequencing (scRNA-seq) analyses to delve into the pathogenesis of bladder ES/PNET. The investigation revealed the presence of specialized types of epithelial cells (referred to as bladder ES-Epi) and mast cells (referred to as bladder ES-Mast) within bladder ES/PNET in comparison to urothelial carcinoma. more...

A multitargeting approach rewires cell identity in fusion-negative rhabdomyosarcoma [bulk_ATACseq]

(Submitter supplied) Rhabdomyosarcoma (RMS), the most common soft-tissue sarcoma occurring in the pediatric population, arises from the skeletal muscle compartment. Several transcriptional and post-transcriptional regulators lock the tumor in a continuous undifferentiated state and prevent the cells from returning to quiescence. MicroRNAs (miRNAs) are short non-coding RNAs that modulate cell identity by means of post-transcriptional regulation of messenger RNAs. more...

Gene expression profiling of trophoblastic organoid from the placental villi of intrauterine and tubal ectopic pregnancies

(Submitter supplied) The tubal ectopic pregnancy (TEP) is a severe pregnancy complication accounted for 10-15% pregnancy-related deaths owing to the implantation and growth of embryos in fallopian tubes. Placental villi from TEP exhibit significant difference compared with that from intrauterine pregnancy (IP) in total volumes, trophoblast differentiation and intravillous vascularization. To investigate the difference in expression profiles of placental villi from IP and TEP, placental villi were collected from each phenotype, separated from maternal tissues and analyzed by high-throughput sequencing. more...

The glucocorticoid receptor elicited proliferative response in human erythropoiesis is BCL11A-dependent, microdeletion patients.

(Submitter supplied) Prior evidence indicates that the erythroid cellular response to glucocorticoids (GC) has developmental specificity, namely, that developmentally more advanced cells that are undergoing or have undergone fetal to adult globin switching are more responsive to GC-induced expansion. To investigate the molecular underpinnings of this, we focused on the major developmental globin regulator BCL11A. We compared: a) levels of expression and nuclear content of BCL11A in adult erythroid cells upon GC stimulation; b) response to GC of CD34+ cells from patients with BCL11A microdeletions and reduced BCL11A expression, and; c) response to GC of two cellular models (HUDEP-2 and adult CD34+ cells) before and after reduction of BCL11A expression by shRNA. more...

ETV5 accelerates therapy-induced neuroendocrine prostate cancer development by inducing neural stem-like cell differentiation [ChIP-seq]

(Submitter supplied) Neuroendocrine prostate cancer (NEPC) is an aggressive subtype induced by hormone therapy, presenting significant therapeutic challenges due to the lack of effective treatments. In this study, we explored the role of ETV5 in NEPC development. Analysis of multiple prostate cancer datasets revealed significantly elevated ETV5 expression in NEPC compared to other subtypes. ETV5 expression increased progressively under hormone therapy conditions through epigenetic modifications. more...

Bulk RNAseq of FNE1 cells with ectopic expression of YAP1 and HPV E6/E7 oncogenes

(Submitter supplied) High grade serous ovarian carcinoma (HGSOC) is the most common and aggressive ovarian malignancy. Accumulating evidence indicates that HGSOC may originate from human fallopian tube epithelial cells (FTECs), although the exact pathogen(s) and/or molecular mechanism underlying the malignant transformation of FTECs is unclear. Here we show that human papillomavirus (HPV), which could reach FTECs via retrograde menstruation or sperm-carrying, interacts with the yes-associated protein 1 (YAP1) to drive the initiation and progression of high grade serous cancer (HGSC). more...

Cell and state-specific eQTL analysis in human endometrial mesenchymal cells uncovers candidate susceptibility genes for reproductive diseases and traits

(Submitter supplied) The functional consequence of genetic variants and how they contribute to disease remains elusive. Studying genetic variants in disease-relevant cells captures variations in effects in cell types and states. The endometrium is a heterogenic tissue with critical reproduction roles considered complex genetic traits. We assessed the association between genetic variants and gene expression in purified endometrial mesenchymal cells using both bulk and single-cell RNA-sequencing data from patients with endometria-driven reproductive disorders. more...

Serial mRNA profiling of peripheral blood monocytes from PAH patients treated with phosphodiesterase 5 inhibitors (PDE5i) and PDE5i+macitentan

(Submitter supplied) Purpose: We performed single cell RNA sequencing (scRNA-seq) of peripheral blood mononuclear cells (PBMCs) from non-diseased control individuals (N=3) as well as from individuals with PAH, naïve to treatment (N=4), 3 months after phosphodiesterase 5 inhibitor (PDE5i) treatment (N=5), and 3 months after treatment with PDE5i plus an endothelin receptor antagonist, macitentan (N=5). We performed complementary analyses of differential gene expression and differential gene network dependency. more...
