Pathway implications of aberrant global methylation in adrenocortical cancer

(Submitter supplied) Context: Adrenocortical carcinomas (ACC) are a rare tumor type with a poor five-year survival rate and limited treatment options. Objective: Understanding of the molecular pathogenesis of this disease has been aided by genomic analyses highlighting alterations in TP53, WNT, and IGF signaling pathways. Further elucidation is needed to reveal therapeutically actionable targets in ACC. Design: In this study, global DNA methylation levels were assessed by the Infinium HumanMethylation450 BeadChip Array on 18 ACC tumors and 6 normal adrenal tissues. more...

Adrenocortical Carcinoma Gene Expression Profiling [Agilent]

(Submitter supplied) Adrenocortical carcinoma (ACC) is an aggressive endocrine tumor with a poor 5 year survival rate of 10-20%. We used expression profiling to assess the transcriptional changes associated with ACC as compared to normal adrenal glands with the goal of identifying new targets for treatment. Fourteen ACC tumors were profiled on both Affymetrix U133 Plus 2 or Agilent 22K Human Genome arrays; an additional six tumors were profiled on one or the other array. more...

Characterization of differential gene expression in adrenocortical tumors harbouring β-catenin (CTNNB1) mutations.

(Submitter supplied) Mutations of β-catenin gene (CTNNB1) are frequent in adrenocortical adenomas (AA) and carcinomas (ACC). However, the target genes of CTNNB1 have not yet been identified in adrenocortical tumors. Our objective was to identify genes de-regulated in adrenocortical tumors harbouring CTNNB1 genetic alterations.

Adrenocortical adenomas harboring β-catenin gene CTNNB1 mutations

Analysis of adrenocortical adenomas (AA) with β-catenin missense mutations and AAs with wild-type CTNNB1. Mutations of CTNNB1 are common in AA and adrenocortical carcinomas (ACC). Results provide insight into molecular mechanisms deregulated in adrenocortical tumors with CTNNB1 genetic alterations.

Human Adrenocortical Tumors: normal cortex vs inactive adenoma vs cortisol secreting adenoma vs adrenocortical cancer

(Submitter supplied) Transcriptional profiling of human adrenocortical tumors. Gene expression profile of normal adrenal cortex, hormonally inactive adenoma, cortisol-secreting Cushing-adenoma and primary adrenocortical cancer tissues were compared. The goal of this study was to identify significant differences in the gene expression of these groups. Further aim was to reveal biologically relevant pathogenetic pathways altered at transcriptional and posttranscriptional level, as well.

Expression profiling of HCC

(Submitter supplied) 61 human HCCs were analyzed for genome-wide gene expression. Samples were collected at two sites in Germany, Heidelberg (HD) and Hannover (N). The Heidelberg Collection include 40 independent HCC: 19 liver resections and 17 explant liver specimen (4 not determined); median age at surgery was 57 years (range, 16-78), and the male/female ratio was 3:1. All diagnoses were confirmed by histological reevaluation, and use of the samples was approved by the local ethics committee. more...

Epigenetic treatment of cervical carcinoma cell lines

(Submitter supplied) Four cervical carcinoma cell lines (C4I, CaSki, HeLa, SiHa) were treated with epigenetic modifying drugs 5-aza-2’-deoxycytidine and trichostatin A. Such treatment leads to reactivation of genes that are epigenetically silenced in cancer by aberrant methylation; reactivated genes were then identified by microarray profiling. Subsets consist of 2 biological repeats and 2 dye-flip experiments for each repeat for CaSki cell line, and 3 repeats, each with 2 dye-flip experiments for C4I, HeLa, Caski cell lines. more...

Gene exrpression profiling of childhood adrenocortical tumors

(Submitter supplied) Pediatric adrenocortical tumors (ACT) are rare and often fatal malignancies; little is known regarding their etiology and biology. To provide additional insight into the nature of ACT, we determined the gene expression profiles of 24 pediatric tumors (five adenomas, 18 carcinomas, and one undetermined) and seven normal adrenal glands. Distinct patterns of gene expression, validated by quantitative real-time PCR and Western blot analysis, were identified that distinguish normal adrenal cortex from tumor. more...

Long non-coding RNA expression profiling reveals LINC00271 as a prognostic marker in adrenocortical carcinoma

(Submitter supplied) Adrenocortical carcinoma (ACC) is an aggressive malignancy with a low but variable overall survival rate. Even after complete tumor resection, over half of patients develop recurrent disease. Long noncoding RNAs (lncRNAs) have been found to be involved in cancer initiation/progression and as markers of cancer prognosis. The role of lncRNAs in ACC is poorly understood. Thus, in this study we performed lncRNA expression profiling in ACC, adrenocortical adenoma (ACA) and normal adrenal cortex (NAC). more...

MicroRNA profiling of benign and malignant adrenocortical tumors reveals potential biomarkers of recurrence

(Submitter supplied) MicroRNAs are small, non-coding RNAs that regulate gene expression at post-transcriptional levels. There is increasing evidence to suggest that miRNAs could be useful in cancer diagnosis, prognosis, and therapy. The aim of our study was to identify miRNAs predictors of poor prognosis in adrenocortical cancer. miRNA microarray expression profiling was performed on a cohort of 6 adenomas, 6 non-recurrent carcinomas (Carc_B) and 6 recurrent carcinomas (Carc_A). more...

Development of MeTA-inducible system in LNCaP cells

(Submitter supplied) Epigenetic gene silencing induced by aberrant DNA methylation is an important mechanism leading to the loss of key cellular pathways in tumorigenesis. Although DNA demethylating agents reverse DNA methylation and have significant cytostatic and cytotoxic effects, it remains elusive whether their effects on cellular phenotypes are solely on reactivation of hypermethylated genes. To address this issue, we constructed LNCaP-derived human prostate cancer cell lines that can pharmacologically induce the expression of fusion gene comprising NFkB transcriptional activation domain (AD) and methyl-CpG binding domain (MBD). more...

A genome-wide search for epigenetically regulated genes in zebra finch using MethylCap-seq and RNA-seq

(Submitter supplied) DNA methylation is tightly linked with gene expression regulation and has long been regarded a stable epigenetic mark in postmitotic cells. However, it recently became clear that postnatal brains appear to show stimulus-induced de novo CpG methylation or active demethylation related to neuronal plasticity. Due to striking homologies between the brains of birds and mammals, songbirds, especially the zebra finch, propose an attractive model for investigating the genome-wide DNA methylation profile and DNA methylation reconfiguration during brain development. more...
