Hand2 loss of function effect on embryonic heart

Analysis of Hand2 mutant embryo hearts at 19 hpf. Hand2 is a myocardial transcription factor essential for fusion of bilateral cardiomyocyte populations moving toward the embryonic midline. Results provide insight into downstream components of the Hand2 pathway that mediates cardiac morphogenesis.

Transcription profile of zebrafish hand2 (hanS6 allele) mutant embryos at 19hpf

(Submitter supplied) Heart formation requires the fusion of bilateral cardiomyocyte populations as they move toward the embryonic midline. The bHLH transcription factor Hand2 is essential for cardiac fusion; however, the effector genes that execute this function of Hand2 are unknown. Here, we provide the first evidence for a downstream component of the Hand2 pathway that mediates cardiac morphogenesis. Although hand2 is expressed in cardiomyocytes, mosaic analysis demonstrates that hand2 plays a non-autonomous role in regulating cardiomyocyte movement. more...

HAND2 Target Gene Regulatory Networks Control Cardiac Valve Development

(Submitter supplied) The HAND2 transcriptional regulator controls cardiac development and we uncover addition essential functions in the endothelial to mesenchymal transition (EMT) underlying cardiac cushion development in the atrioventricular canal (AVC). In Hand2-deficient mouse embryos, the EMT underlying AVC cardiac cushion formation is disrupted and we combined ChIP-Seq of embryonic hearts with transcriptome analysis of wild-type and mutants AVCs to identify the functionally relevant HAND2 target genes. more...

Expression data from Hand2 mutant mouse embryos

(Submitter supplied) Acquisition of the lower jaw (mandible) was evolutionarily important for jawed vertebrates. In humans, syndromic craniofacial malformations often accompany jaw anomalies. Hand2 is involved in coordinating the developmental network of mandibles and the oral apparatus through Hand2-downstream genes and is therefore a major determinant of jaw identity. We used microarrays to detail the global programme of gene expression involved in mandible specification and identified distinct classes of up-regulated genes during this process.

Zebrafish her3 knockout impacts developmental and cancer-related gene signatures

(Submitter supplied) HES3 is a basic helix-loop-helix transcription factor that regulates neural stem cell renewal during development. HES3 overexpression is predictive of reduced overall survival in patients with fusion-positive rhabdomyosarcoma, a pediatric cancer that resembles immature and undifferentiated skeletal muscle. However, the mechanisms of HES3 cooperation in fusion-positive rhabdomyosarcoma are unclear and are likely related to her3/HES3’s role in neurogenesis. more...
