Two distinct Trypanosoma eIF4F complexes co-exist, bind different mRNAs and are regulated during nutritional stress

(Submitter supplied) Many eIF4F subunits and PABP paralogues are found in trypanosomes: six eIF4E, five eIF4G, one eIF4A and two PABPs. They are expressed simultaneously and assemble into different complexes, contrasting the situation in metazoans that use distinct complexes in different cell types or developmental stages. Each eIF4F complex has its own proteins, mRNAs and, consequently, a distinct function. We set out to study the function and regulation of the two major eIF4F complexes of Trypanosoma cruzi. more...

Experimental and Bioinformatic upgrade for genome-wide mapping of nucleosomes in Trypanosoma cruzi

(Submitter supplied) We adapted the widely used digestion of chromatin with micrococcal nuclease (MNase) followed by deep sequencing to the parasite Trypanosoma cruzi, which presents numerous singularities. In this work, we use the hybrid CL Brener strain carrying two set of chromosomes from two substantially different parental strains. The hybrid strain CL Brener is composed of the Esmeraldo-like and non Esmeraldo-like haplotypes. more...

Gene expression profiling of Trypanosoma cruzi in the presence of heme points to glycosomal metabolic adaptation of epimastigotes inside the vector

Differential transcriptome of human dendritic cells at first contact with Trypanosoma cruzi reveals response to virus as a central unexplored pathway

(Submitter supplied) Chagas disease is a notorious public health problem worldwide. However, the pathology is still poorly understood in humans and dendritic cells (DCs) have a determinant role during infection that could help to understand what happens with immune response during the first contact. Here we performed a transcriptomic analysis of the interaction between DCs from three human donors and the infective forms of T. more...

Global changes in nitration levels and DNA binding profile of Trypanosoma cruzi histones induced by incubation with host extracellular matrix

(Submitter supplied) Adhesion of T. cruzi trypomastigotes to components of the extracellular matrix (ECM) is an important step in mammalian host cell invasion. We have recently described a significant increase in the tyrosine nitration levels of histones H2A and H4 when trypomastigotes are incubated with components of the ECM. In this work, we used chromatin immunoprecipitation (ChIP) with an anti-nitro-tyrosine antibody followed by mass spectrometry to identify nitrated DNA binding proteins in T. more...

Ribonomic identification of mRNAs associated with RNA binding proteins TcRBP7 and TcRBP40 from Trypanosoma cruzi

(Submitter supplied) TcRBP7 and TcRBP40 proteins are adjacent paralogs in T. cruzi with a conserved RRM structure and similar protein localization at specific foci at the posterior region. We identified their targets in order to compare their function, infering about the evolutionary implication of regulatory proteins duplication. Surprisingly, the mRNA targets co-immunoprecipitated with each RBP were distinct, pointing to a subfunctionalization outcome. more...
